      目的回顾性分析中心体相关蛋白 55(CEP55)、睾丸表达基因 14(TEX14)在结直肠癌组织中表达及其临床意义。方法选取 2015年 10月至 2016年 10月在南阳市中心医院行手术治疗的 78例结直肠癌病人癌组织及其癌旁组织为研究对象。利用 Ualcan数据库分析 CEP55和 TEX14在结直肠癌组织及癌旁组织中的表达情况;实时荧光定量 PCR(qRT-PCR)法检测 CEP55和 TEX14 mRNA表达;免疫组化染色法检测 CEP55和 TEX14蛋白表达;分析 CEP55和 TEX14蛋白表达与病人临床病理特征的关系;对病人随访 5年,使用 Kaplan-Meier法绘制生存曲线,分析 CEP55和 TEX14蛋白表达与结直肠癌病人 5年生存率的关系;使用 Cox回归模型分析影响结直肠癌病人预后的因素。结果 Ualcan数据库分析结果显示,结直肠癌组织中 CEP55和 TEX14表达量均明显高于癌旁组织( P<0.05)。结直肠癌组织中 CEP55、TEX14 mRNA水平( 2.34±0.70比 1.00±0.26、1.57±0.44比0.98±0.25)及蛋白阳性表达率均明显高于癌旁组织( P<0.05)。CEP55和 TEX14蛋白表达与分化类型、有无淋巴结转移、 TNM分期、有无神经浸润、有无肝转移有关( P<0.05)。结直肠癌组织中 CEP55蛋白阳性病人生存率为 52.73%,明显低于 CEP55蛋白阴性病人生存率 78.26%(P<0.05); TEX14蛋白阳性病人生存率为 52.63%,明显低于 TEX14蛋白阴性病人生存率80.95%(P<0.05)。低分化、淋巴结转移、 TNM分期 Ⅲ~Ⅳ期、 CEP55蛋白阳性、 TEX14蛋白阳性是影响结直肠癌病人 5年预后的独立危险因素( P<0.05)。结论 CEP55和 TEX14在结直肠癌组织中呈高表达, CEP55和 TEX14蛋白阳性病人 5年生存率低于阴性病人,这些指标的临床意义,值得进一步研究。

      Objective The expression and clinical significance of centrosome-associated protein 55 (CEP55) and testis expressed gene 14 (TEX14) in colorectal cancer tissues were analyzed retrospectively.Methods Seventy-eight cases of colorectal cancer patientswho underwent surgical treatment in Nanyang Central Hospital from October 2015 to October 2016 were selected as the research objects. The expressions of CEP55 and TEX14 in colorectal cancer and adjacent tissues were analyzed by Ualcan database; the mRNA expressions of CEP55 and TEX14 were detected by quantitative real-time PCR (qRT-PCR); the expressions of CEP55 and TEX14 proteins were detected by immunohistochemical staining; the relationship between the expression of CEP55 and TEX14 proteins and clinicopathological features of patients was analyzed; the patients were followed up for 5 years, the survival curve was drawn by Kaplan-Meier method, and the relationship between the expression of CEP55 and TEX14 protein and the 5-year survival rate of colorectal cancerpatients was analyzed; Cox regression model was used to analyze the factors affecting the prognosis of patients with colorectal cancer.Results The results of Ualcan database analysis showed that the expressions of CEP55 and TEX14 in colorectal cancer tissues weresignificantly higher than those in adjacent tissues (P<0.05). The mRNA levels and protein positive expression rates of CEP55 andTEX14 in colorectal cancer were significantly higher than those in adjacent tissues (2.34±0.70 vs. 1.00±0.26, 1.57±0.44 vs. 0.98±0.25) (P<0.05). The expression of CEP55 and TEX14 protein was related to differentiation type, lymph node metastasis, TNM stage, nerve invasion and liver metastasis (P<0.05). The survival rate of patients with positive CEP55 protein in colorectal cancer was 52.73 %, whichwas significantly lower than that of CEP55 negative patients 78.26% (P<0.05); the survival rate of patients with TEX14 protein positivewas 52.63%, which was significantly lower than that of patients with TEX14 protein negative was 80.95% (P<0.05). Low differentiation, lymph node metastasis, TNM stage Ⅲ-Ⅳ, CEP55 protein positive and TEX14 protein positive were independent risk factors affecting the 5-year prognosis of colorectal cancer patients (P<0.05).Conclusions CEP55 and TEX14 are highly expressed in colorectal cancer. The 5-year survival rate of CEP55 and TEX14 protein positive patients is lower than that of negative patients, the clinical significance of these indexes is worthy of further study.
