      目的探讨长链非编码 RNA(LncRNA)人浆细胞瘤转化迁移基因 1()、微 RNA-31-5p(miR-31-5p)在慢性乙型病毒性肝炎(CHB)病人血浆中的表达情况,并分析其与炎症损伤程度的关系。方法选取东莞市人民医院 2019年 12月至 2020年 12月确诊的 180例 CHB病人为 CHB组,所有病人均行肝穿刺活检,根据 CHB病人炎症损伤程度分为 G1组(66例),G2组(58例),G3(41例)G4组(15例)根据肝纤维化程度进行分组,无明显肝纤维化(S0,S1)组(64例)明显肝纤维化组(S2,S3)(78例),早期组肝硬化(S4,)组(38例)。,同时选取 55例健康者为对照组。收集 CHB组和对照组的一般资,料,分别检测两组血清肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-6、IL-1β、IL-10、超敏 C反应蛋白(Hs-CRP)、丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST)、谷氨酰基转移酶(GGT)等指标水平;采用实时荧光定量 PCR法对血清中 LncRNA PVT1、miR-31-5p表达水平进行检测,Pearson法分析 LncRNA PVT1、miR-31-5p表达的相关性。结果 CHB病人血清中 LncRNA PVT1(1.52±0.31、1.93±0.47、2.57±0.63、 3.42±1.03)表达水平高于对照组 0.68±0.23,且 LncRNA PVT1表达水平随着炎症损伤程度加重而升高,miR-31-5p表达水平(0.85±0.22、0.79±0.18、0.63±0.14、0.50±0.11)低于对照组 0.96±0.24,且 miR-31-5p表达水平随着炎症损伤程度加重而降低,差异有统计学意义(P<0.05);CHB组病人 TNF-α(108.13±30.32)ng/L、IL-6(68.50±22.59)ng/L、IL-1β(358.92±36.80)ng/L、IL-10(75.25±18.99)ng/L、Hs-CRP(11.22±3.15)mg/L、ALT(57.13±9.34)U/L、AST(37.86±5.56)U/L、GGT水平(33.21±10.62)U/L均显著高于对照组[(38.49±19.88)ng/L、(19.12±3.21)ng/L、(169.05±5.24)ng/L、(16.56±2.34)ng/L、(1.68±0.56)mg/L、(24.62±7.95)U/L、(22.32±4.26)U/L、(26.94±8.32)U/L](P<0.05);CHB组病人血清中 LncRNA PVT1表达与 TNF-α、IL-6、IL-1β、IL-10、Hs-CRP、 ALT、AST、GGT均呈正相关(P<0.05)miR-31-5p与 TNF-α、IL-6、IL-1β、IL-10、Hs-CRP、ALT、AST、GGT均呈负相关(P<0.05); CHB病人血清中 LncRNA PVT1、miR-315p的表达呈明显负相关(r=.0.36,P<0.05)。结论 CHB病人血清中 LncRNA PVT1表达升高,miR-31-5p表达降低,且二者均与炎症损伤程度有关。

      Objective To investigate the expressions of long non-coding RNA (LncRNA) transforming migration gene 1 (PVT1) and microRNA-31-5p (miR-31-5p) in the plasma of patients with chronic hepatitis B (CHB) virus infection, and to analyze their relationshipwith the degree of inflammatory injury.Methods A total of 180 CHB patients diagnosed in Dongguan People's Hospital from Decem.ber 2019 to December 2020 were selected as the CHB group. All patients underwent liver biopsy and were assigned into G1 group (n= 66), G2 group (n=58), G3 group (n=41) and G4 group (n=15) according to the degree of inflammatory injury of CHB patients. And theywere also assigned into no obvious liver fibrosis (S0, S1) group (n=64), obvious liver fibrosis group (S2, S3) n=78), and early cirrhosis (S4) group (n=38) according to the degree of liver fibrosis. At the same time, 55 healthy subjects were gathered as the control group.General data of the CHB group and the control group were collected, and serum tumor necrosis factor-α (TNF-α), interleukin (IL)-6, IL-1β, and IL-10, high-sensitivity C-reactive protein (Hs-CRP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), glutam.yltransferase (GGT) and other index levels were measured. The expression levels of LncRNA PVT1 and miR-31-5p in serum were mea. sured by real-time fluorescence quantitative PCR, and the correlation of LncRNA PVT1 and miR-31-5p expression was analyzed byPearson method.Results The expression levels of LncRNA PVT1 in the serum of CHB patients were higher than that in the control group [(1.52±0.31, 1.93±0.47, 2.57±0.63, 3.42±1.03) vs. (0.68±0.23)], and the expression level of LncRNA PVT1 increased with the ag.gravation of inflammatory injury. The expression levels of miR-31-5p were lower than that in the control group [(0.85±0.22, 0.79±0.18,0.63±0.14, 0.50±0.11) vs. (0.96±0.24)], and the expression level of miR-31-5p reduced with the aggravation of inflammatory injury (P< 0.05). The levels of TNF-α, IL-6, IL-1β, IL-10, Hs-CRP, ALT, AST and GGT in the CHB group were obviously higher than those in the control group [(108.13±30.32) ng/L vs. (38.49±19.88) ng/L, (68.50±22.59) ng/L vs. (19.12±3.21) ng/L, (358.92±36.80) ng/L vs. (169.05± 5.24) ng/L, (75.25±18.99) ng/L vs. (16.56±2.34) ng/L, (11.22±3.15) mg/L vs. (1.68±0.56) mg/L, (57.13±9.34) U/L vs. (24.62±7.95) U/L,(37.86±5.56) U/L vs. (22.32±4.26) U/L, (33.21±10.62) U/L vs. (26.94±8.32) U/L] (P<0.05). The expression level of LncRNA PVT1 in serum of CHB group was directly correlated with the levels of TNF-α, IL-6, IL-1β, IL-10, Hs-CRP, ALT, AST and GGT (P<0.05), while the expression level of miR-31-5p was inversely correlated with the levels of TNF-α, IL-6, IL-1β, IL-10, Hs-CRP, ALT, AST and GGT (P<0.05). The expressions of LncRNA PVT1 and miR-31-5p in serum of CHB patients were obviously inversely correlated (r=.0.36, P< 0.05).Conclusion The expression of LncRNA PVT1 in serum of CHB patients is increased, and the expression of miR-31-5p is re. duced. Both are related to the degree of inflammatory damage.
