      目的比较柴胡提取物及其所含皂苷 a和皂苷 d促进骨骼增长的作用及其机制研究。方法自 2021年 3月至 2022年 1月进行了高效液相色谱法测定柴胡提取物中柴胡皂苷 a与 d含量;然后将昆明种小鼠 40只采用随机数字表法分为健康对照组、柴胡皂苷 a组(简称皂苷 a组)、柴胡皂苷 d组(简称皂苷 d组)、柴胡提取物组(简称提取物组)和阳性对照组,每组 8只。健康对照组灌胃去离子水,皂苷 a组灌胃柴胡皂苷 a 1.15 μg/g体质量,皂苷 d组灌胃柴胡皂苷 d 0.2 μg/g体质量,柴胡提取物组按照 0.94 mg/g体质量灌胃,阳性对照组灌胃酸枣仁皂苷 a0.013 mg/g体质量灌胃。灌胃 25 d后于末次给药 30 min后取脑组织,蛋白质印迹法检测脑组织色氨酸羟化酶 2(TPH2)含量, ELISA法检测脑组织 5-羟色胺( 5-HT)、生长激素( GH)含量的变化和 5-HT与 5-羟色胺 1A受体( 5-HT1AR)结合活性,并测量各组药物对小鼠胫骨长度的影响。结果柴胡提取物中柴胡皂苷 a含量为 1.22 mg/g,柴胡皂苷 d含量为 0.22 mg/g;提取物组和皂苷 a组的脑组织 5-HT含量( 60.93±11.09,59.51±11.02)μg/L、GH含量(68.91±9.10,68.42±9.03)μg/L、5-HT1AR与 5-HT结合活性( 39.90±8.36,37.27±6.59)及胫骨长度( 10.56±0.53,10.55±0.54)cm均较健康对照组[(40.89±8.74)μg/L,(44.87±8.92)μg/L,27.34±3.45,(9.48±0.63)cm]明显增高( P<0.01),同时这两组 TPH2含量较健康对照组升高( P<0.01)均差异有统计学意义。皂苷 d组脑组织 TPH2蛋白含量、 5-HT含量( 47.46±8.62)、 GH含量( 45.81± 7.14)、 5-HT1AR与 5-HT结合,活性( 30.87±5.52)及胫骨长度( 9.88±0.56)cm较健康对照组差异无统计学意义( P>0.05)。与皂苷 a组比较,皂苷 d组的脑组织 TPH2蛋白含量、 5-HT含量、 5-HT1AR与 5-HT结合活性及胫骨长度减少( P<0.05)GH含量明显减少( P<0.01),差异有统计学意义。与皂苷 d组比较,提取物组 GH含量、 5-HT1AR与 5-HT结合活性及胫骨长度增,加明显增高(P<0.01),5-HT含量增高( P<0.05),差异有统计学意义。结论柴胡提取物延缓慢波睡眠时长的主要成分为柴胡皂苷 a,其可

      Objective To compare the role and mechanism of bupleurum extract, saikosaponin a and saikosaponin d in promoting bone growth.Methods The contents of saikosaponin a and d in bupleurum extract were determined by high-performance liquid chro-matography from March 2021 to January 2022. Forty Kunming strain mice were randomly assigned into normal control group, saikosa-ponin a group (referred to as saponin a group), saikosaponin d group (referred to as saponin d group), bupleurum extract group (referredto as extract group) and positive control group. Normal control group received gavage with deionized water, saponin a group with saiko-saponin a 1.15 μg/g body weight, saponin d group gavage saikosaponin d 0.2 μg/g body weight, bupleurum extract group 0.94 mg/gbody weight, and positive control group acid jujube kernel saponin a 0.013 mg/g body weight. After 25 days of gavage, brain tissue ofmice was taken 30 minutes after the last dose, and western blotting was used to detect the content of tryptophan hydroxylase 2 (TPH2)in brain tissue, and ELISA was used to detect the change of 5-hydroxytryptamine (5-HT) and growth hormone (GH) in brain tissue, and the binding activity of 5-HT and 5-hydroxytryptamine 1A receptor (5-HT1AR), and the effect of drugs in each group on the tibia length of mice was also measured.Results The contents of saikosaponin a in the extract was 1.22 mg/g, while that of saikosaponin d was 0.22 mg/g. The 5-HT and GH contents of brain tissue , binding activity of 5-HT1AR and 5-HT and tibial length in the extract group and thesaponin a group were significantly increased [(60.93±11.09, 59.51±11.02)μg/L, (68.91±9.10, 68.42±9.03)μg/L, (39.90±8.36, 37.27±6.59) and (10.56±0.53, 10.55±0.54) cm] in comparison with those in normal control group [(40.89±8.74)μg/L, (44.87±8.92)μg/L,(27.34±3.45), and (9.48±0.63) cm] (P<0.01). Meanwhile, the contents of TPH2 in these two groups were also increased in comparison with normal control group (P<0.01), and differences were found to be significant. The contents of TPH2, 5-HT (47.46±8.62), and GH (45.81±7.14) in brain tissue, binding activity of 5-HT1AR and 5-HT (30.87±5.52) and tibial length (9.88±0.56) of mice in the saponind group were not statistically different from those in normal control group (P>0.05). Compared with saponin a group, the tibial length, the contents of TPH2 and 5-HT, and the binding activity of 5-HT1AR and 5-HT were reduced in the saponin d group (P<0.05), and the GH content in the saponin d group was significantly reduced (P<0.01); differences were found to be significant. Compared with saponin d group, the GH content, binding activity of 5-HT1AR and 5-HT and tibial length of the extract group increased significantly (P<0.01), and the content of 5-HT also increased (P<0.05); differences were found to be significant.Conclusions Saikosaponin a is the main component in saikosaponin extract that increases the duration of slow-wave sleep. The possible mechanism is that saikosaponin a in-creases the high expression of tryptophan hydroxylase protein in brain tissue, thereby promoting the secretion of 5-HT, enhancing the binding activity of 5-HT1AR and 5-HT, prolonging the duration of slow-wave sleep, increasing the secretion of growth hormone, and promoting bone growth in mice.
