      目的探究多囊卵巢综合征( PCOS)病人血清 Ⅲ型纤连蛋白域蛋白 5(FNDC5)、过氧化物酶体增殖物激活受体 γ辅助激活因子 -1α(PGC-1α)表达水平与胰岛素抵抗的关系。方法选取 2017年 1月至 2021年 12月在重庆松山医院接受治疗的 118例 PCOS病人为 PCOS组,另选取与 PCOS病人年龄、身体质量指数( BMI)、腰臀比相匹配的同期健康体检者 94例作为对照组。酶联免疫吸附测定( ELISA)检测两组血清 FNDC5、PGC-1α表达水平;全自动生化分析仪检测空腹胰岛素( FINS)、空腹血糖(FBG)水平,并计算胰岛素抵抗相关指标胰岛 β细胞功能指数( HOMA-β)和胰岛素抵抗指数( HOMA-IR); Pearson相关性分析研究病人血清 FNDC5、PGC-1α水平与胰岛素抵抗相关指标之间的关系。结果与对照组相比, PCOS组 FBG、FINS、三酰甘油(TG)、 HOMA-β、HOMA-IR显著升高( P<0.05);与对照组相比, PCOS组 FNDC5[( 18.46±2.76)μg/L比( 30.25±5.87)μg/L]、 PGC1α[(1.87±0.45)μg/L比( 4.91±1.34)μg/L]表达水平显著降低( P<0.05)。 Pearson相关性分析结果显示, PCOS病人血清 FNDC5、 PGC-1α水平均与 FBG、FINS、HOMA-β、HOMA-IR呈显著负相关( P<0.05)。结论 PCOS病人血清 FNDC5、PGC-1α表达水平降低,与胰岛素抵抗存在负相关。

      Objective To investigate the relationship between serum levels of fibronectin domain Ⅲ protein 5 (FNDC5) and peroxi some proliferator-activated receptor-γ coactivator factor 1α (PGC-1α) and insulin resistance in patients with polycystic ovarian syn drome (PCOS).Methods From January 2017 to December 2021, 118 patients with PCOS admitted to the Department of Endocrinology in Songshan General Hospital were selected as the PCOS group. Another 94 healthy subjects matching the age, body mass index(BMI) and waist-to-hip ratio of PCOS patients during the same period were selected as the control group. The expression levels of FNDC5 and PGC-1α in serum of the two groups were detected by enzyme-linked immunosorbent assay (ELISA). The levels of fasting insulin (FINS) and fasting blood glucose (FBG) were detected by automatic biochemical analyzer, and homeostasis model assessment forβ cell function (HOMA-β) and homeostasis model assessment of insulin resistance (HOMA-IR) were calculated. Pearson correlation was used to analyze the relationship between serum FNDC5 and PGC-1α levels and insulin resistance-related indicators. Results Compared with the control group, FBG, FINS, triglyceride (TG), HOMA-β and HOMA-IR in the PCOS group were obviously increased (P<0.05). Compared with the control group, the expression levels of FNDC5[(18.46±2.76) μg/L vs. (30.25±5.87) μg/L] and PGC-1α [(1.87±0.45) μg/L vs. (4.91±1.34) μg/L] in the PCOS group were obviously decreased (P<0.05). Pearson correlation analysis showed that serum FNDC5 and PGC-1α level in PCOS patients was obviously negatively correlated with FBG, FINS, HOMA-β, and HOMA-IR (P<0.05).Conclusion The expression levels of serum FNDC5 and PGC-1α in PCOS patients are decreased, which is negatively corre lated with insulin resistance.
