      目的观察增液八珍汤对慢传输型便秘大鼠结肠匀浆及血清中超氧化物歧化酶( SOD)、谷胱甘肽( GSH)及一氧化氮(NO)的影响。方法 2022年 7—11月将由徐州医科大学实验动物中心供应的 72只 SPF级 Wistar大鼠按随机数字表法分为六组,每组 12只,包括空白组( A),模型组(B)、麻仁软胶囊组( C)、增液八珍汤低( D)、中( E)、高剂量组( F)。 C组给予麻仁软胶囊内容物: 6.2 mL·kg-1·day-1D、E、F组分别给予 17、34、68 g·kg-1·d-1的增液八珍汤灌胃,共给药 14 d,A、B组给等量的 0.9%氯化钠溶液。末次给药后,检测各,组大鼠血清和结肠匀浆中 SOD、GSH、NO的含量。结果增液八珍汤低剂量组血清 SOD、GSH,结肠组织 SOD、GSH表达水平[( 2.90±0.28)μmol/L、(8.43±1.56)μmol/L、(0.93±0.32)μmol/L、(6.62±0.48)μmol/L]、中剂量组[( 4.02±0.55)μmol/L、(11.75±2.46)μmol/L、(1.76±0.25)μmol/L、(8.92±0.86)μmol/L]、高剂量组[( 4.67±0.86)μmol/L、(13.43±1.49)μmol/L、(1.84±0.60)μmol/L、(10.32±1.64)μmol/L]相较于模型组[( 1.98±0.21)μmol/L、(5.78±1.98)μmol/L、(0.56±0.47) μmol/L、(4.36±1.58)μmol/L]均明显升高( P<0.05)。增液八珍汤低、中、高剂量组血清 NO、结肠组织 NO表达水平相较于模型组均明显降低( P<0.05)。与 B组比较, C、D、E、F组均能明显增加便秘大鼠血清和结肠匀浆中 SOD、GSH的含量; E、F组增加便秘大鼠血清和结肠匀浆中 SOD、GSH优于 C、D组。与 B组比较, C、D、E、F组均能明显减少便秘大鼠血清和结肠匀浆中 NO的含量; E、F组减少便秘大鼠血清和结肠匀浆中 NO优于 C、D组。结论增液八珍汤能够通过调节便秘大鼠 SOD、GSH及 NO在血液及肠壁组织中的含量水平,达到减少过氧化反应,减轻氧自由基及一氧化氮对机体的损害,达到促胃肠动力的作用。

      Objective To observe the effects of Zengye Bazhen decoction on superoxide dismutase (SOD), glutathione (GSH) and nitric oxide (NO) in colonic homogenate and serum of rats with slow-transit constipation.Methods Seventy-two rats provided by theLaboratory Animal Center of Xuzhou Medical University from July 2022 to November 2022 were randomly assigned into blank group(A), model group (B), Maren soft capsule group (C), low-dose group (D), medium-dose group (E), and high-dose group (F) of Zengye Ba- zhen decoction. Group C was given Maren soft capsule contents: 6.2 mL·kg-1·day-1, and groups D, E and F were given 17 g·kg-1·d-1,34 g·kg-1·d-1, 68 g·kg-1·d-1 of Zengye Bazhen decoction respectively for gastric gavage for a total of 14 days, and groups A and Bwere given the same amount of normal saline. After the last dosing, the contents of SOD, GSH and NO in the serum and colonic homogenate of rats in each group were detected.Results The expression levels of serum SOD and GSH and SOD and GSH in colon tissues in low-dose group [(2.90±0.28) μmol/L, (8.43±1.56) μmol/L, (0.93±0.32) μmol/L, (6.62±0.48) μmol/L], medium-dose group [(4.02±0.55) μmol/L, (11.75±2.46) μmol/L, (1.76±0.25) μmol/L, (8.92±0.86) μmol/L], and high dose group of Zengye Bazhen decoction [(4.67±0.86) μmol/L, (13.43±1.49) μmol/L, (1.84±0.60) μmol/L, (10.32±1.64)] were significantly increased (P<0.05) in comparison with those in the model group [(1.98±0.21) μmol/L, (5.78±1.98) μmol/L, (0.56±0.47) μmol/L, (4.36±1.58) μmol/L]. The expression levels of NO in serum and colon tissue in low-dose, medium-dose and high-dose groups of Zengye Bazhen decoction were significantly lower than those in the model group (P<0.05). The contents of SOD and GSH in serum and colonic homogenate of constipatedrats were significantly increased in both groups C, D, E and F(P<0.05 compared with group B), the increase of SOD and GSH in serumand colonic homogenate of rats in groups E and F was better than that in groups C and D. The contents of NO in serum and colonic homogenate of rats with constipation were significantly reduced in groups C, D, E and F (P<0.05 compared with group B). The reductionof NO in serum and colonic homogenate of rats in groups E and F was better than that in groups C and D.Conclusion Zengye Ba- zhen decoction can reduce peroxidation by adjusting the contents of SOD, GSH and NO in blood and intestinal wall tissues in constipated rats, and alleviate the damage of oxygen free radicals and nitric oxide to the body, thus achieving the effect of promoting gastrointestinal motility.
