目的探讨泛素样蛋白质 2(UBQLN2)在结直肠癌中的表达及其对结直肠癌细胞增殖的影响。方法选取 2020年 1月至 2021年 1月河北北方学院附属第一医院治疗的 90例病人为研究对象,根据术中手术获取组织情况,分为正常组、癌旁组和肿瘤组。免疫组织化学法和免疫荧光评估 UBQLN2在肿瘤组织、远端组织和正常组织中表达;分析 UBQLN2与病人临床病理特征以及预后的相关性;蛋白质印迹法和实时荧光定量逆转录聚合酶链反应( qRT-PCR)检测 UBQLN2在结直肠癌细胞系 HT29、SW620、SW480、LOVO、HCT116以及人正常肠上皮细胞 HIEC细胞中的表达;分别过表达和干扰 UBQLN2在结肠癌细胞中的表达,随后 CCK-8检测干扰前后细胞增殖能力变化。结果 UBQLN2在结直肠癌癌旁组织中阳性面积( 32.57±12.44)%显著高于肿瘤组织( 12.68±9.79)%;UBQLN2 mRNA表达在转移阴性病人肿瘤组织中 0.98±0.15显著高于转移阳性病人肿瘤组织 |

Objective To evaluate the expression of ubiquilin 2 (UBQLN2) gene in colorectal cancer and its role in colorectal cancercell proliferation and invasion.Methods The study included 90 patients who underwent treatment in The First Affiliated Hospital ofHebei North University from January 2020 to January 2021. Based on intraoperative tissue acquisition, the patients were assigned intothree groups: normal group, para-carcinoma group, and tumor group. Immunohistochemistry and immunofluorescence techniques wereused to detect the expressions of UBQLN2 in tumor tissue. distal tissue and normal tissue. The correlation between UBQLN2 expres-sion, clinicopathological features and prognosis was analyzed. Real-time fluorescent quantitative reverse transcription polymerase chain reaction (qRT-PCR) and Western blotting were performed to measure UBQLN2 expression levels in HT29, SW620, SW480, LOVO,HCT116 and human normal intestinal epithelial cells (HIEC cells), respectively. After overexpression and interference of UBQLN2 incolon cancer cells, CCK-8 assays were used to assess cell proliferation before and after interference.Results The postive area rate of UBQLN2 in the para-carcinoma tissues of colorectal cancer was significantly higher than that in the tumor tissues [(32.57±12.44) % vs. (12.68±9.79) %]. The mRNA expression level of UBQLN2 in tumor tissues from metastasis-negative patients was significantly higher than that observed in tumor tissues from metastasis-positive patients (0.98±0.15 vs. 0.33±0.08). Moreover, the expression of UBQLN2 was positively correlated with patient prognosis. Analysis using qRT-PCR and Western blotting showed a higher expression of UBQLN2in HIEC cells than in colorectal cancer cells and lowest in HCT116 cells. Thirty-six hours after UBQLN2 overexpression in HCT116 cells, the proliferation capacity of pcDNA3.1-UBQLN-2 group cells was significantly reduced compared to that of the pcDNA3.1-NC group cells (0.52±0.18 vs. 0.78±0.23). Thirty-sxi hours after UBQLN2 knockdown in SW480 cells, the proliferation capacity of siUBQLN-2 group cells was significantly enhanced compared to that of si-NC cells (0.89±0.36 vs. 0.65±0.37).Conclusions UBQLN2expression is downregulated in colorectal cancer tissues and cells and negatively correlated with metastasis. It can inhibit the prolifera-tion of colorectal cancer cells and is positively correlated with patient prognosis. |
