目的探究 ERCC6L蛋白在胃癌组织中的表达变化及其与临床病理特征和预后的关系。方法选取南通大学附属启东医院 2015年 4月至 2017年 5月行手术治疗的 150例胃癌病人为研究对象,收集胃癌组织及癌旁非肿瘤组织,采用实时荧光定量逆转录 PCR(RT-qPCR)及免疫组织化学法检测 ERCC6L mRNA及蛋白表达水平,分析其与临床病理特征关系;采用 Kaplan-Meier法分析 ERCC6L蛋白表达与病人预后的关系;采用 Cox回归分析影响胃癌病人预后的相关因素。结果胃癌组织中 ERCC6L mRNA的相对表达量 1.60±0.53显著高于癌旁非肿瘤组织 1.01±0.12(t=13.30,P<0.001)、 ERCC6L蛋白阳性表达率均显著高于癌旁非肿瘤组织( P<0.05)。 ERCC6L蛋白表达与淋巴结转移、肿瘤浸润深度、 TNM分期、远处转移、分化程度有关(P<0.05)。胃癌组织 ERCC6L阳性表达病人 5年总生存率( 19.57%)显著低于 ERCC6L阴性表达病人( 60.34%,P<0.05)。 ERCC6L阳性表达、淋巴结转移、 TNMⅢ~TNMⅣ和低分化均是影响胃癌病人不良预后的独立危险因素( P<0.05)。结论 ERCC6L在胃癌组织中高表达,与分化程度、肿瘤浸润深度、淋巴结转移、 TNM分期、远处转移及预后密切相关,是影响胃癌病人预后的独立危险因素。 |

Objective To investigate the change in expression of ERCC6L protein in gastric cancer (GC) and its relationship withclinicopathological characteristics and prognosis.Methods A total of 150 GC patients who underwent surgical treatment in QidongHospital affiliated to Nantong University from April 2015 to May 2017 were selected as the study subjects, GC tissues and non-tumor tissues adjacent to cancer were collected, RT-qPCR and immunohistochemistry were applied to detect the expression levels of ERCC6LmRNA and protein, the relationship between clinical and pathological features was analyzed; Kaplan-Meier method was applied to analyze the relationship between ERCC6L protein expression and prognosis; Cox regression analysis was applied to analyze the related factors affecting the prognosis of GC patients.Results The relative expression of ERCC6L mRNA in GC tissue was significantly higher at 1.60 ± 0.53 compared to 1.01 ± 0.12 in paracancerous non tumor tissues (t=13.30, P <0.001). The positive expression rate of ERCC6Lprotein in GC tissues was obviously higher than that in paracancerous non tumor tissues (P<0.05). ERCC6L protein expression was correlated with lymph node metastasis, tumor invasion depth, TNM stage, distant metastasis and differentiation (P<0.05). The 5-year overall survival rate of patients with ERCC6L positive expression in GC tissue (19.57%) was obviously lower than that of patients with ERCC6L negative expression (60.34%, P<0.05). ERCC6L positive expression, lymph node metastasis, TNM Ⅲ-TNM Ⅳ and low differentiation were independent risk factors for poor prognosis of GC patients (P<0.05).Conclusions ERCC6L is highly expressed in GC tissues, which is closely related to the degree of differentiation, depth of tumor invasion, lymph node metastasis, TNM stage, distant metastasis and prognosis, and is an independent risk factor affecting the prognosis of GC patients. |
