目的探究慢性宫颈炎合并人乳头状瘤病毒( HPV)感染病人血清性别决定区 Y框蛋白 2(SOX2)和程序性死亡配体 1(PD-L1)表达与临床病理特征相关性。方法选取 2020年 10月至 2022年 10月在重庆市江津区中医院就诊治疗的未合并 HPV感染慢性宫颈炎病人 160例作为慢性宫颈炎组,慢性宫颈炎合并 HPV感染病人 140例作为合并 HPV组,另选取在该院体检健康女性 110例作为对照组。采用酶联免疫吸附测定( ELISA)检测研究对象血清 SOX2水平,实时荧光定量 PCR(qRT-PCR)测定血清 PD-L1 mRNA水平,二代杂交捕获实验技术( HC2)检测 HPV-DNA。结果对照组、慢性宫颈炎组、合并 HPV组血清 SOX2、PD-L1mRNA水平呈现逐渐升高的趋势( P<0.05);低危型 HPV组、中危型 HPV组、高危型 HPV组血清 SOX2、PD-L1 mRNA水平呈现逐渐升高的趋势( P<0.05);慢性宫颈炎合并 HPV病人中单一感染率为 45.71%(64/140)多重感染率为54.29%(76/140)(P>0.05),其中慢性宫颈炎合并 HPV病人中低危型单一感染与多重感染病例数差异有统计学意,义( P<0.05);高水平 SOX2与低水平 SOX2以及高水平 PD-L1 mRNA与低水平 PD-L1 mRNA慢性宫颈炎病人合并 HPV率差异有统计学意义( P<0.05)。结论慢性宫颈炎合并 HPV感染病人血清 SOX2、PD-L1 mRNA水平均显著升高,与 HPV分型密切相关,且病人血清 SOX2、PD-L1 mRNA水平与 HPV感染均呈正相关。 |

Objective To investigate the correlation between the expression of serum sex-determining region of Y chromosome-related high-mobility-group box 2 (SOX2), programmed death ligand-1 (PD-L1) in patients with chronic cervicitis complicated with humanpapillomavirus (HPV) infection and the clinicopathological characteristics.Methods A total of 160 cases of chronic cervicitis without HPV infection treated in Jiangjin District Hospital of Traditional Chinese Medicine from October 2020 to October 2022 were selectedas the chronic cervicitis group, and 140 cases of chronic cervicitis with HPV infection were selected as the HPV group, another 110 healthy women who underwent physical checkup in our hospital were selected as the control group. The serum SOX2 level of the subjects was measured by enzyme-linked immunosorbent assay (ELISA), real-time fluorescence quantitative PCR (qRT-PCR) was applied to detect the level of PD-L1 mRNA in serum, and the HPV-DNA was detected by second-generation hybridization capture assay (HC2). Results The levels of serum SOX2 and PD-L1 mRNA in control group, chronic cervicitis group and HPV group showed a gradually increasing trend (P<0.05). The levels of serum SOX2 and PD-L1 mRNA in low-risk HPV group, medium-risk HPV group and high-risk HPV group showed a gradually increasing trend (P<0.05). The single infection rate in patients with chronic cervicitis complicated withHPV was 45.71% (64/140), and the multiple infection rate was 54.29% (76/140) (P>0.05). There was a statistically significant difference in the numbers of single infection and multiple infection in patients with chronic cervicitis complicated with HPV (P<0.05). Therewere significant differences in the rate of HPV in patients with chronic cervicitis between high SOX2 level and low SOX2 level, and between high PD-L1 mRNA level and low PD-L1 mRNA level.Conclusion The levels of serum SOX2 and PD-L1 mRNA in patientswith chronic cervicitis complicated by HPV infection are obviously increased, which is closely related to HPV typing, and the levels ofserum SOX2 and PD-L1 mRNA are positively correlated with HPV infection. |
