目的使用双样本孟德尔随机化( MR)分析肠道菌群与肝硬化的因果关系,阐明个体基因组特征对精准干预病人肠道菌群以改善肝硬化的可能性。方法该研究于 2023年 9―12月获取肠道微生物群等位基因单核苷酸多态性( SNP)(13 266例样本)的工具变量及肝硬化相关等位基因 SNP(1 011例欧洲病例和 356 077例欧洲对照样本)的结局数据,使用 MR分析肠道菌群对肝硬化的因果关系。结果随机效应下的逆方差加权( IVW)分析结果表明,放线菌属[P=0.001,OR=1.39,95%CI:(1.13,1.71)]普雷沃菌属[P=0.017,OR=1.27,95%CI:(1.04,1.54)]与肝硬化呈正相关。甲烷短杆菌属[P=0.024,OR=0.86,95%CI:(0.75, 8)],乳酸菌属[P=0.042,OR=0.82,95%CI:(0.68,0.99)]与肝硬化呈负相关。结论放线菌属、普雷沃菌属与肝硬化0.9,存在因果关系,具有正面促进效应。 |

Objective To analyze the causal relationship between the gut microbiota and cirrhosis by using two-sample mendelianrandomization (MR) analysis, and to elucidate the potential impact of individual genomic characteristics on interventions targeting thegut microbiota to improve cirrhosis precisely. Methods Instrumental variables for gut microbiota allele single nucleotide polymorphism (SNP) (13 266 samples) and outcome data for cirrhosis-related allele SNP (1 011 European samples and 356 077 European control samples) were obtained from September to December 2023. Mendelian randomization (MR) analysis was employed to test the causalrelationship between gut microbiota and cirrhosis.Results The inverse variance weighted (IVW) analysis results indicated that Actino? myces [P=0.001,OR=1.39,95%CI:(1.13,1.71)],Prevotella 9[P=0.017,OR=1.27,95%CI:(1.04,1.54)] were positively associated with cirrhosis. Methanobrevibacter [P=0.024,OR=0.86,95%CI:(0.75,0.98)], Lactobacillus [P=0.042,OR=0.82,95%CI:(0.68,0.99)] were negatively associated with cirrhosis.Conclusion There is a causal relationship between Actinomyces, Prevotella 9 genera and cirrhosis, with a positive promoting effect. |
