目的研究霍山石斛多糖(DHP)对帕金森病( PD)模型神经炎性损伤的作用。方法 2022年 6月至 2023年 2月,将小胶质细胞 BV-2分为正常组(不做任何干预正常培养)、模型组[以 1-甲基 -4-苯基吡啶离子( MPP+)诱导]、 DHP组(先行 2~400 mg/L DHP干预,后加模型组相同剂量 MPP+诱导),应用酶联免疫吸附测定( ELISA)检测细胞白细胞介素( IL)-1β含量; 30只 C57BL/6小鼠采用随机数字表法分为正常组、模型组、 DHP组,每组 10只。设置 DHP等效治疗剂量即 350 mg/kg为 DHP组。造模完成后,正常组和模型组灌胃双蒸水 0.5 mL,1次/天,持续 2周; DHP组灌胃 0.5 mL DHP溶液,浓度为 15.4 g/mL,1次/天,持续 2周。通过平衡木实验和转棒试验观察小鼠平衡能力、肌力以及运动协调力; ELISA检测小鼠中脑组织 IL-1β和 IL-18炎症因子表达水平;免疫荧光染色检测小鼠中脑黑质离子钙结合衔接分子 1(Iba-1)和黑质酪氨酸羟化酶(TH)表达水平。结果实验结果显示,与正常组比较, BV-2细胞模型组 IL-1β水平显著升高[(76.32±6.18)ng/L比( 51.12±4.72)ng/L,P<0.05];与模型组比较, DHP组 IL-1β水平降低( P<0.05)且在 2.0~100.0 mg/L内呈剂量依赖性下降。与正常组比较,模型组通过平衡木时间显著延长( P< 0.05),在棒时间缩短( P<0.05中脑组织 IL-1β[(190.10±14.68)ng/L比( 80.46±15.18)ng/L]和 IL-18水平升高( P<0.05);与模型组比较, DHP组通过平衡木运间缩短(P<0.05),在棒时间延长(P<0.05)DHP组中脑组织 IL-1β[(89.69±14.31)ng/L]和 IL-18水平降低(P<0.05)。免疫荧光结果显示,与正常组比较,模型组中脑黑质 Iba1表达显著升高(P<0.05)TH表达下降( P<0.05);与模型组比较, DHP组中脑黑质 Iba-1表达显著降低( P<0.05),TH表达升高( P<0.05)。结论 DHP通过抑制小胶质细胞),动时,-,可能,过度活化,进而降低 PD模型神经炎性损伤,发挥神经保护作用。 |

Objective To investigate the neuroprotective effects of polysaccharides from Dendrobium huoshanense (DHP) on neuroin-flammatory damage in a Parkinson's disease (PD) model.Methods BV-2 microglial cells were divided into three groups: normal con-trol (NC, no intervention for normal culture), model (Mod, Induced with MPP+), and DHP-treated (first 2 to 400 mg/L DHP intervention, plus the same dose of MPP+ induction in the model group) group. The levels of interleukin-1β (IL-1β) in cells were measured using en-zyme-linked immunosorbent assay (ELISA). Thirty C57BL/6 mice were randomly assigned to three groups (n=10 per group): NC, Mod,and DHP. The DHP group received an equivalent dose of 350 mg/kg DHP. After modeling, the NC and Mod groups received daily ga-vage with 0.5 mL ddH2O for two weeks, while the DHP group received 0.5 mL of a 15.4 g/L DHP solution daily for two weeks. Balancebeam and rotarod tests were used to assess the mice's balance, muscle strength, and motor coordination. Levels of inflammatory cyto-kines IL-1β and IL-18 in midbrain tissue were measured by ELISA, while immunofluorescence staining was used to evaluate expres-sion levels of ionized calcium-binding adapter molecule 1 (Iba-1) and tyrosine hydroxylase (TH) in the substantia nigra.Results Com-pared with the NC group, IL-1β levels in the BV-2 cell model group were significantly increased [(76.32±6.18) ng/L vs. (51.12±4.72) ng/ L, P<0.05]. In comparison with the Mod group, IL-1β levels in the DHP group decreased in a dose-dependent manner within the 2.0-100.0 mg/L range (P<0.05). Compared to the NC group, the Mod group showed a significant increase in balance beam time and a reduc-tion in rotarod time (P<0.05), with elevated levels of IL-1β [(190.10±14.68) ng/L vs. (80.46±15.18) ng/L] and IL-18 in midbrain tissue (P<0.05). Compared to the Mod group, the DHP group demonstrated a reduction in balance beam time (P<0.05), an increase in rotarod time (P<0.05), and decreased IL-1β [(89.69±14.31) ng/L] and IL-18 levels(P<0.05). Immunofluorescence results showed that compared with the NC group, Iba-1 expression in the substantia nigra of the Mod group was significantly increased (P<0.05), while TH expression decreased (P<0.05). Compared to the Mod group, the DHP group showed a significant reduction in Iba-1 expression and an increase in TH expression (P<0.05).Conclusion DHP may exert neuroprotective effects by inhibiting excessive activation of microglia, therebyreducing neuroinflammatory damage in PD models. |
