      目的观察牛血清白蛋白 -硫化铜( BSA-CuS)联合光热治疗( PTT)对糖尿病足溃疡( DFU)小鼠氧化应激、炎症反应的影响。方法 2023年 1―5月,运用化学法制备 BSA-CuS,并处理培养的金黄色葡萄球菌、大肠杆菌,平板涂布法计算抑菌率。链脲佐菌素腹腔注射联合创面混合菌涂布法制造 DFU小鼠模型,分别给予 BSA-CuS给药、 PTT治疗、 BSA-CuS联合 PPT治疗。酶联免疫吸附试验( ELISA)检测血清中丙二醛( MDA)、超氧化物歧化酶( SOD)、人白细胞介素( IL)-1、IL-6和肿瘤坏死因子 α(TNF-α)的含量;蛋白质印迹法检测创面组织中无翼型 MMTV聚集家族成员 3A(Wnt3a)、 β-连环蛋白( β-catenin)、糖原合酶激酶-3β(GSK-3β)蛋白表达。结果与磷酸缓冲盐溶液( PBS)+PPT组相比, BSA-CuS+PPT组大肠杆菌[(56.48±6.54)%比( 6.48±1.37)%]、金黄色葡萄球菌[( 71.47±7.71)%比( 6.56±1.38)%]的抑菌率均明显升高( P<0.05)。 BSA-CuS联合 PPT处理相较于 BSA-CuS或 PPT单独处理, DFU小鼠的创面愈合率升高( P<0.05)MDA、IL-1、IL-6和 TNF-α均显著降低( P<0.05),SOD显著升高( P<0.05)Wnt3a、β-catenin蛋白表达均显著升高( P<0.05)GSK-3,β蛋白表达显著降低( P<0.05)。结论 BSA-CuS联合 PPT可抑制 DFU小,鼠体内的氧化应激和炎症反应,利于创面愈合,其,机制可能与失活 Wnt信号通路有关。
英文摘要:
      Objective To observe the effect of bovine serum albumin-CuS (BSA-CuS) combined with photothermal therapy (PTT) on oxidative stress and inflammatory response of mice with diabetic foot ulcers. Methods BSA-CuS nanomaterials were prepared by chemical method, and cultured StaphylococCuS aureus and Escherichia coli were treated from January to May 2023. The inhibition ratewas calculated by plate coating method. DFU mice model was established by intraperitoneal injection of streptozotocin combined withwound mixed bacteria coating. BSA-CuS, PTT and BSA-CuS-PTT were given respectively. The content of malondialdehyde (MDA), su-peroxide dismutase (SOD), interleukin (IL)-1, IL-6 and tumor necrosis factor α (TNFα) in serum were detected by enzyme linked immu-nosorbent assay (ELISA). Western blotting (WB) was used to detect the protein expressions of Wingless type MMTV integration sitefamily member 3A (Wnt3a), β-catenin, and glycogen synthase kinase-3 β (GSK-3β) in wound tissue.Results Compared with phos-phate buffer saline (PBS) + PTT group, the inhibition rates of Escherichia coli [(56.48±6.54) % vs. (6.48±1.37) %] and Staphylococcus aureus [(71.47±7.71) % vs. (6.56±1.38) %] in BSA-CuS+PTT group were significantly increased (P<0.05). Compared with BSA-CuS or PTT alone, the wound healing treated with BSA-CuS combined with PTT was significantly increased (P<0.05), and the levels of MDA, IL-1, IL-6 and TNF-α were significantly decreased (P<0.05). And meanwhile, SOD was significantly increased (P<0.05), the protein ex-pressions of Wnt3a, β-Catenin were significantly increased (P<0.05), while GSK-3β expression was significantly decreased (P<0.05) in DFU mice.Conclusion BSA-CuS combined with PTT could inhibit the oxidative stress and inflammatory reaction of DFU, which isbeneficial to wound healing, and its mechanism may be related to the inactivation of Wnt signaling pathway.
