目的探究苦参碱对子宫肌瘤细胞凋亡和自噬的影响。方法将原代子宫肌瘤细胞分为 3组,对照组利用常规培养基培养,低剂量组在常规培养基中加入 0.5 g/L苦参碱,高剂量组加入 2 g/L苦参碱。分别利用流式细胞仪和多核苷酸链断裂检测技术检测细胞凋亡水平,利用免疫荧光技术检测细胞中自噬斑点,利用投射电镜检测细胞中自噬体,利用蛋白质印迹法检测凋亡及自噬相关蛋白含量。结果对照组相对凋亡比为 1.00±0.02,低剂量组相对凋亡比为 2.97±0.13,高剂量组相对凋亡比为 3.18±0.17,与对照组相比,低剂量组和高剂量组细胞凋亡增加(t低剂量组比对照组 =26.07,P低剂量组比对照组 =1.286×10.5;t高剂量组比对照组 =22.73, P高剂量组比对照组 =2.219×10.5)而低剂量组和高剂量组细胞凋亡差异无统计学意义(t高剂量组比低剂量组 =1.73,P高剂量组比低剂量组 =0.158);对照组自噬细胞比例为 3.33±0.70,,低剂量组自噬细胞比例为 53.44±5.61,高剂量组自噬细胞比例为 56.51±6.48。与对照组相比,低剂量组和高剂量组自噬荧光斑点细胞比例增加(t低剂量组比对照组 =15.36,P低剂量组比对照组 =1.048×10.4;t高剂量组比对照组 =14.12,P高剂量组比对照组 = |

Objective To explore the effect of matrine on the apoptosis and autophagy of uterine fibroid cells.Methods The primaryuterine fibroid cells were assigned into control group, low-dose group and high-dose group. Cells in the control group were cultured inconventional medium, cells in the low dose group and high dose group were treated with 0.5 g/L and 2 g/L matrine, respectively. Flowcytometry and polynucleotide chain break detection technology were used to detect the apoptosis level, the immunofluorescence tech-nology to detect autophagic spots in cells, transmission electron microscope to detect autophagosomes, and Western blotting to detectthe levels of proteins related to apoptosis and autophagy.Results The relative apoptosis ratios of the control group, the low-dose groupand the high-dose group were 1.00±0.02, 2.97±0.13, and 3.18±0.17, respectively. Compared with the control group, the apoptosis ofcells in the low-dose group and high-dose group increased (tlow-dose group vs. control group=26.07, Plow-dose group vs. control group=1.286×10.5; thigh-dose group vs. control group =22.73, Phigh-dose group vs. control group=2.219×10.5), while there was no statistically significant difference in cell apoptosis between the low-dose group and high-dose group (thigh-dose group vs. low-dose group=1.73, Phigh-dose group vs. low-dose group=0.158). The proportions of autophagic cells in the control group, the low-dose group and the high-dose group were 3.33±0.70,53.44±5.61,and 56.51±6.48, respectively. Compared with the con-trol group, the proportions of autophagic fluorescent spot cells increased in the low-dose group and high-dose group (tlow-dose group vs. control group= 15.36, P low-dose group vs. control group=1.048×10.4; thigh-dose group vs. control group=14.12, Phigh-dose group vs. control group=1.458×10.4), while there was no statistically sig-nificant difference between the low-dose group and the high-dose group (thigh-dose group vs. Low-dose group=0.62, Phigh-dose group vs. Low-dose group=0.569).Conclu. sion Martine can enhance apoptosis and autophagy of uterine fibroid cells. |
