      目的探究黄芪多糖对溃疡性结肠炎大鼠结肠病理损伤及辅助性 T细胞 17/调节性 T细胞( Th17/Treg)免疫平衡的影响。方法于 2022年 1月至 2023年 1月,将 60只大鼠按照随机数字表法分为对照组( NC组)、模型组( UC组),黄芪多糖低、中、高剂量组( HQDT-L组、 HQDT-M组、 HQDT-H组)和阳性对照组( SASP组)。观察大鼠一般情况;评估 DAI评分; ELISA检测血清中白细胞介素 6(IL-6)、白细胞介素 10(IL-10)含量;检测大鼠髓过氧化物酶( MPO)活性;苏木精 -伊红( HE)染色检测结肠组织病理学变化;流式细胞仪检测大鼠脾脏组织 Treg/Th17比值;蛋白质印迹法检测结肠组织中维甲酸相关孤儿受体( ROPR-γt)和叉头框蛋白 P3(FoxP3)蛋白表达。结果干预前, NC组大鼠 DAI评分为 0分,其余各组大鼠 DAI评分明显升高( P<0.001)且各组间差异无统计学意义( P>0.05);干预后 UC组大鼠 DAI评分( 2.87±0.56)分、血清中 IL-6(25.61±10.83)ng/L含量、MPO活性,(2.36±0.28)、结肠组织病理学评分( 2.83±0.21)分、 Th17细胞百分比( 65.32±5.46)%及 RORγt蛋白( 0.21±0.03)表达明显高于 NC组,血清中 IL-10(6.89±0.74)ng/L含量、 Treg细胞百分比( 6.94±1.05)%和 Treg/Th17比值( 0.12±0.03)、 FoxP3蛋白(0.15±0.02)表达明显低于 NC组( P<0.001);和 UC组相比, HQDT-L组、 HQDT-M组和 HQDT-H组大鼠 DAI评分( 2.16±0.32)分、(1.54±0.26)分、(1.12±0.17)分,血清中 IL-6含量( 201.34±8.15)ng/L、(141.57±7.42)ng/L、(75.32±6.85)ng/L,MPO活性( 2.01±0.21、1.46±0.15、0.65±0.07),结肠组织病理学评分( 2.24±0.19)分、(1.76±0.15)分、(1.08±0.12)分, Th17细胞百分比( 52.11±4.32)%、(45.81±3.16)%、(31.26±2.54)%及 RORγt蛋白( 0.17±0.02、0.14±0.02、0.10±0.01)表达明显降低,血清中 IL-10含量(8.36±0.81)ng/L、(10.27±0.94)ng/L、(13.81±1.05)ng/L,Treg细胞百分比( 10.32±1.42)%、(17.61±1.53)%、(22.45±1.76)%和 Treg/ Th17比值( 0.21±0.04、0.38±0.06、0.72±0.08)、 FoxP3蛋白( 0.31±0.04、0.55±0.06、0.69±0.07)表达明显升高( P<0.001); HQDT-H组和 SASP组大鼠各指标相比差异无统计学意义( P>0.05)。结论黄芪多糖对溃疡性结肠炎大鼠结肠病理损伤的改善作用,可能和调节 Th17/Treg免疫平衡、 RORγt/FoxP3表达水平抑制炎症反应相关。

      Objective To investigate the effect of Astragalus polysaccharides on colonic pathological damage and immune balance ofhelper T cells 17/regulatory T cells (Th17/Treg) in rats with ulcerative colitis.Methods From January 2024 to January 2025, 60 ratswere randomly divided into a control group (NC group), a model group (UC group), low, medium, and high dose groups of Astragaluspolysaccharides (HQDT-L group, HQDT-M group, HQDT-H group), and a positive control group (SASP group) according to a randomnumber table method. the general condition of rats was observed; DAI score was evaluated; ELISA was used to detect the levels of inter-leukin-6 (IL-6) and interleukin-10 (IL-10) in serum; the activity of rat myeloperoxidase (MPO) was detected; Hematoxylin eosin (HE)staining was used to detect pathological changes in colon tissue; Flow cytometry was used to detect the Treg/Th17 ratio in rat spleen tis-sue; Western blot was used to detect retinoic acid associated orphan receptor (ROPR -γ t) and forkhead box protein P3 (FoxP3) protein expression in colon tissue.Results Before intervention, the DAI score of rats in the NC group was 0, while the DAI scores of rats inthe other groups increased significantly (P<0.001), and there was no statistically significant difference between the groups (P>0.05). Af-ter intervention, the DAI score (2.87±0.56) points, IL-6 content (25.61±10.83) ng/L, MPO activity (2.36±0.28), colon histopathologicalscore (2.83±0.21) points, Th17 cell percentage (65.32±5.46)%, and ROR γ t protein expression (0.21±0.03) in the UC group were sig-nificantly higher than those in the NC group. The IL-10 content (6.89±0.74) ng/L, Treg cell percentage (6.94±1.05), Treg/Th17 ratio (0.12±0.03), and FoxP3 protein expression (0.15±0.02) in the serum were significantly lower than those in the NC group (P<0.001). Compared with the UC group, rats in the HQDT-L group, HQDT-M group, and HQDT-H group had DAI scores of (2.16±0.32) points, (1.54±0.26) points, (1.12±0.17) points, serum IL-6 levels of [(201.34±8.15) ng/L, (141.57±7.42) ng/L, and (75.32±6.85) ng/L], MPO ac-tivity (2.01±0.21, 1.46±0.15, 0.65±0.07), colon histopathological score [(2.24±0.19) points, (1.76±0.15) points, (1.08±0.12) points],Th17 cell percentage [(52.11±4.32)%, (45.81±3.16)%, (31.26±2.54)%], and ROR γ t protein expression (0.17±0.02, 0.14±0.02, 0.10±0.01) were significantly reduced. Serum IL-10 content [(8.36±0.81) ng/L, (10.27±0.94) ng/L, (13.81±1.05) ng/L], Treg cell percentage[(10.32±1.42)%, (17.61±1.53) %, (22.45±1.76)%]. The Treg/Th17 ratio (0.21±0.04, 0.38±0.06, 0.72±0.08) and the expression of FoxP3protein (0.31±0.04, 0.55±0.06, 0.69±0.07) were significantly increased (P<0.001). There was no statistically significant difference in various indicators between HQDT-H group and SASP group rats (P>0.05).Conclusion The improvement effect of astragalus polysac-charide on colonic pathological damage in rats with ulcerative colitis may be related to the regulation of Th17/Treg immune balance,RORγt/FoxP3 expression levels, and inhibition of inflammatory reactions.
