      目的:探讨HMGB-1、P-AKT和MMP-9在大鼠急性痛风性关节炎滑膜和关节软骨表达及意义。方法 将清洁级的雄性Sprague-Dawley大鼠随机分为A组(实验组)和B组(对照组),两组均在自由摄取诱导饲料(3%氧嗪+15%酵母+82%基础饲料)和饮用正常水的基础下,连续饲养6周;实验组,将MSU结晶混悬液0.2mL(25mg/mL)经US介导下注射到大鼠右侧踝关节间隙,对照组为相同方法注射0.2mL生理盐水。造模期间,定时测量大鼠血尿酸值变化、步态变化、肾脏组织病理切片,并对滑膜与关节软骨进行组织病理学观察与免疫组织化学染色评估HMGB-1、P-AKT、MMP-9的表达水平。结果 与对照组相比较,实验组SD大鼠踝关节肿胀度更高(P<0.05)、步态跛行变化更加明显,各时间点HMGB-1、P-AKT、MMP-9的表达水平更高 。结论 HMGB-1、P-AKT、MMP-9在SD大鼠AGA滑膜及关节软骨表达水平更高,可能与其参与炎症反应及介导免疫因子的释放有关。

      Objective To investigate the expression and significance of HMGB-1, P-AKT and MMP-9 in articular cartilage and synovium in rats with acute gouty arthritis.Methods Clean-grade male Sprague-Dawley rats were randomly divided into group A(experimental group )and group B(control group). Both groups were re-fed for 6 weeks on the basis of free intake of induction diet (3% oxazine, 15% yeast, 82% basal diet) and drinking normal water. The experimental group injected 0.2ml (25mg/mL) of the MSU crystal suspension into the right ankle joint space of the rats under US mediation, and the control group was injected with 0.2mL normal saline by the same method. During the modeling period, the changes in blood uric acid value, body weight, general condition, skin temperature, joint swelling, gait changes, and renal histopathological sections were measured regularly, and the expression levels of HMGB-1, P-AKT, MMP-9 were evaluated by histopathological observation and immunohistochemical staining of articular cartilage and synovium.Results Compared with the control group(P<0.05), the SD rats in the experimental group had higher disease activity index scores, more obvious changes in gait claudication, higher ankle swelling, and higher expression levels of HMGB-1, P-AKT, MMP-9 at each time point.Conclusion The expression levels of HMGB-1, P-AKT and MMP-9 in AGA articular cartilage and synovium are higher in SD rats, which may be related to their involvement in inflammatory response and mediating the release of immune factors.
