目的探讨胃癌病人癌组织中信号转导及转录激活蛋白 3(STAT3)、葡萄糖 -6-磷酸脱氢酶(G6PD)表达水平及其与临床病理特征和病人预后的关系。方法收集 2018年 3月至 2019年 8月在海南西部中心医院进行治疗的 96例胃癌病人作为研究对象,根据病人预后 3年生存情况分为生存组 62例,死亡组 34例。收集病人胃癌组织样本和癌旁组织样本(距胃癌组织 >5 cm)采用免疫组织化学法检测 STAT3、G6PD表达,分析 STAT3、G6PD表达与病人临床病理特征间关系,实时荧光定量逆转录聚合酶,链反应( qRT-PCR)检测 STAT3 mRNA、G6PD mRNA相对表达水平,采用 Kaplan-Meier法分析 STAT3 mRNA和 G6PD mRNA表达与病人 3年生存率的关系,采用多因素 Cox回归分析病人预后影响因素。结果胃癌组织 STAT3 85.42%(82/96)、 G6PD 87.50%(84/96)蛋白阳性表达率显著高于癌旁组织[ 23.96%(23/96)、 20.83%(20/96)](P<0.05);胃癌组织 STAT3 mRNA 1.24±0.31、G6PD mRNA 1.32±0.39相对表达水平显著高于癌旁组织( 1.02±0.18、0.97±0.16)(P<0.05); STAT3和 G6PD蛋白表达与肿瘤分化程度、临床分期和淋巴结转移显著相关( P<0.05);生存组和死亡组肿瘤分化程度、临床分期、淋巴结转移、 STAT3和 G6PD mRNA表达差异有统计学意义( P<0.05); STAT3、G6PD mRNA低表达病人生存率显著高于高表达病人;临床分期、淋巴结转移、分化程度、 STAT3、G6PD mRNA表达均为胃癌病人预后影响因素( P<0.05)。结论胃癌组织中 STAT3和 G6PD表达升高,与病人临床病理特征和预后有关,可能参与胃癌的发生发展。 |

Objective To investigate the expression levels of signal transducer and activator of transcription 3 (STAT3) and glucose-6-phosphate dehydrogenase (G6PD) in the tissues of patients with gastric cancer and their relationship with clinicopathological character-istics and prognosis.Methods Ninety-six patients with gastric cancer who were treated in Hainan West Central Hospital from March2018 to August 2019 were selected as the study subjects, the patients were grouped into survival group (62 cases) and death group (34cases) according to their 3-year survival. Gastric cancer tissue samples and paracancerous tissue samples were collected from patients(>5 cm from gastric cancer tissue), immunohistochemical method was applied to detect the expression of STAT3 and G6PD proteins,the relationship between the expression of STAT3 and G6PD proteins and the clinicopathological characteristics of patients was ana-lyzed, the relative expression levels of STAT3 mRNA and G6PD mRNA were detected by real-time fluorescent quantitative PCR (qRT-PCR), Kaplan-Meier method was applied to analyze the relationship between the relative expression level of STAT3 mRNA and G6PDmRNA and the 3-year survival rate of gastric cancer patients, multivariate Cox regression was applied to analyze the prognostic factorsof gastric cancer patients.Results The positive expression rates of STAT3 85.42% (82/96) and G6PD 87.50% (84/96) proteins in gas-tric cancer tissue were obviously higher than those in adjacent tissues [23.96% (23/96), 20.83% (20/96)] (P<0.05); the relative expres-sion levels of STAT3 mRNA 1.24±0.31 and G6PD mRNA 1.32±0.39 were obviously higher than those in adjacent tissues (1.02±0.18,0.97±0.16)(P<0.05); the expression of STAT3 and G6PD proteins in gastric cancer was obviously correlated with tumor differentiation,clinical stage and lymph node metastasis (P<0.05); there were statistically obvious differences between survival group and death groupin tumor differentiation, clinical stage, lymph node metastasis, STAT3 mRNA and G6PD mRNA expression (P<0.05); the survival rateof patients with low STAT3, and G6PD mRNA expression was obviously higher than that of patients with high expression; clinical stage,lymph node metastasis, degree of differentiation, expression of STAT3 mRNA and G6PD mRNA were the prognostic factors of gastriccancer patients (P<0.05).Conclusion The expression levels of STAT3 and G6PD in gastric cancer tissue are increased, which are re-lated to the clinicopathological characteristics and prognosis of gastric cancer patients, and may be involved in the development of gas-tric cancer. |
