      目的探讨多囊卵巢综合征( PCOS)病人血清 FOXO1和 FOXO3表达与性激素及胰岛素抵抗的关系。方法选取桂林医学院第二附属医院 2022年 5月至 2023年 5月收治的 85例 PCOS病人(研究组)为研究对象,另选取因非激素或非月经异常就诊的 72例妇女为对照组,空腹采集静脉血,酶联免疫吸附分析( ELISA)测定血清 FOXO1和 FOXO3水平;以氧化电极法、化学发光法测定空腹血糖、空腹胰岛素水平;使用化学发光分析仪测量性激素指标 -总睾酮、黄体生成素、雌二醇水平;以 Pear-son法分析血清 FOXO1和 FOXO3水平相关性及二者与胰岛素抵抗指数及性激素水平。结果研究组生殖内分泌异常的病人占比高于对照组( P<0.05),而在年龄、身体质量指数、饮酒史、代谢异常、吸烟史等方面比较差异无统计学意义( P>0.05);与对照组相比,研究组空腹胰岛素水平[(21.22±2.11)mU/L比( 8.44±0.86)mU/L]、空腹血糖水平[(5.88±0.62)mmol/L比( 4.43±0.52) mmol/L]、胰岛素抵抗指数( 5.45±0.59比 1.66±0.21)、总睾酮[( 1.52±0.17)mU/L比( 1.08±0.12)mU/L]、黄体生成素[( 8.86±0.91) IU/L比( 6.56±0.68)IU/L]、雌二醇[( 138.28±13.94)pmol/L比( 101.55±11.66)pmol/L]、血清[ FOXO1(420.44±45.51)mg/L比(357.24±35.88)mg/L]和 FOXO3[(882.42±90.21)ng/L比( 750.22±78.36)ng/L]水平显著增加(P<0.05); Pearson法分析显示, PCOS病人血清中 FOXO1和 FOXO3水平呈现正相关性,二者分别与总睾酮、胰岛素抵抗指数、雌二醇、黄体生成素呈现正相关性( P<0.05)。结论 PCOS病人血清 FOXO1和 FOXO3表达上调,分别与性激素及胰岛素抵抗具有正相关性。

      Objective To explore the relationship between serum FOXO1 and FOXO3 expressions, sex hormones, and insulin resis-tance in patients with polycystic ovary syndrome (PCOS).Methods A total of 85 patients with PCOS (disease group) admitted to TheSecond Affiliated Hospital of Guilin Medical University from May 2022 to May 2023 were selected as the study subjects, and 72 womenwho sought medical help due to non hormonal or non menstrual abnormalities as the control group. Venous blood was collected on anempty stomach, and enzyme-linked immunosorbent assay (ELISA) was applied to measure serum levels of FOXO1 and FOXO3. The lev-els of fasting blood glucose and fasting insulin were measured by oxidation electrode method and chemiluminescence method. Chemilu-minescence analyzer was applied to measure the levels of total testosterone, luteinizing hormone and estradiol; Pearson correlation coef-ficient was applied to analyze the correlation between serum FOXO1 and FOXO3 levels, and their relationship with insulin resistanceindex and sex hormone levels.Results The proportion of patients with reproductive endocrine abnormalities in the study group was higher than that in the control group (P<0.05), but there was no statistically obvious difference in age, body mass index, history of alco-hol consumption, metabolic abnormalities, and smoking history (P>0.05). Compared with the control group, levels of the fasting insulin [(21.22±2.11) mU/L vs. (8.44±0.86) mU/L], fasting blood sugar [(5.88±0.62) mmol/L vs. (4.43±0.52) mmol/L], insulin resistance index (5.45±0.59 vs. 1.66±0.21), total testosterone [(1.52±0.17) mU/L vs. (1.08±0.12) mU/L], luteinizing hormone [(8.86±0.91) IU/L vs. (6.56± 0.68) IU/L], estradiol [(138.28±13.94) pmol/L vs. (101.55±11.66) pmol/L], serum FOXO1 [(420.44±45.51) mg/L vs. (357.24±35.88) mg/ L] and FOXO3 [(882.42±90.21) ng/L vs. (750.22±78.36) ng/L] in the study group were obviously increased (P<0.05). Pearson correla-tion coefficient analysis results showed that the levels of FOXO1 and FOXO3 in the serum of PCOS patients were positively correlatedwith each other, and they were also positively correlated with total testosterone, insulin resistance index, estradiol, and luteinizing hor-mone, respectively (P<0.05).Conclusion The up-regulated expressions of FOXO1 and FOXO3 in serum of PCOS patients are posi-tively correlated with sex hormones and insulin resistance, respectively.
