| 目的探究防己饮片与防己配方颗粒之间的化学成分变化,旨在为防己配方颗粒的质量控制和应用提供科学依据。方法 2023年 1月至 2024年 2月通过超高效液相色谱与四极杆飞行时间质谱联用技术( UPLC-Q-TOF/MS)明确了防己饮片和防己配方颗粒中的化学成分。此外,运用主成分分析( PCA)和正交偏最小二乘法判别分析( OPLS-DA)方法,进行了多元统计分析,以鉴定防己饮片和配方颗粒主要化学成分及其差异性。结果鉴定了 17种主要生物碱类化合物作为防己饮片和防己配方颗粒共有成分,包括 3种苄基异喹啉类生物碱、 4种阿朴啡类生物碱、 4种四氢原小檗类生物碱、 1种原卟啉类生物碱和 5种双苄基异喹啉类生物碱。通过 PCA分析,证实了样本在化学成分上的总体差异性; OPLS-DA进一步确认了防己饮片与配方颗粒在成分含量分布上的显著区分,并筛选出去甲异波尔定碱、异紫堇定碱和千金藤碱作为差异化学成分。结论防己饮片与配方颗粒在化学成分上表现出一致性及差异性,尤其是差异化合物如:去甲异波尔定碱、异紫堇定碱和千金藤碱的识别有助于确 |

| Objective To explore the chemical composition changes between Stephania tetrandra radix decoction pieces and formulagranules, aiming to provide a scientific basis for their quality control and application of STR formula granules.Methods Ultra-high Performance Liquid Chromatography-Quadrupole Time-of-Flight Mass Spectrometry (UPLC-Q-TOF/MS) was used to determine thechemical composition of STR decoction pieces and formula granules from January 2023 to February 2024. Multivariate statistical analy-ses, including Principal Component Analysis (PCA) and Orthogonal Partial Least Squares-Discriminant Analysis (OPLS-DA), were uti-lized to identify the major chemical components and their differences in STR decoction pieces and formula granules. Results This study successfully identified 17 common alkaloid chemical components in both STR decoction pieces and formula granules, including 3benzylisoquinoline alkaloids, 4 aporphine alkaloids, 4 tetrahydroprotoberberine alkaloids, 1 protoporphyrin alkaloid and 5 dibenzyliso-quinoline alkaloids. PCA analysis was used to confirm the overall difference in chemical composition of the samples. OPLS-DA further confirmed the significant difference in component content distribution between Stephania tetrandra radix decoction pieces and formulagranules, and meisopoldine, isocorydaline and cepharanthine were screened out as differential chemical components.Conclusion The chemical compositions of STR decoction pieces and formula granules showed consistency and differences, especially the identificationof differential compounds such as demethyl isopole alkaloid, isopurpurin alkaloid, and gibberellin alkaloid and it helps to ensure theconsistency and reliability of its therapeutic efficacy, which is of great guiding significance for improving the quality control standardsof STR formula granules. |
