      目的:本研究旨在通过构建小鼠结直肠腺瘤模型,探讨补阳还五汤对小鼠结直肠腺瘤(CRA)生长的抑制作用,并深入分析其与转化生长因子-β1(TGF-β1)/Smad信号通路的关系。方法:将10只野生型C57BL/6J小鼠为正常组,40只Apcmin/+小鼠按照随机数字表法分为4组,每组10只:模型组、低剂量组、高剂量组和激动剂组。观察和记录各组小鼠结直肠腺瘤的数量。HE染色观察结直肠病理变化。检测结直肠组织中IL-6、TNF-α和MCP-1含量。免疫组化检测结直肠组织COX-2和PEG2表达。运用蛋白免疫印迹技术,对TGF-β1/Smad信号通路中相关蛋白的表达水平展开检测。结果:与正常组相比,模型组小鼠结直肠组织可见大量腺瘤,结直肠组织的IL-6 、TNF-α和MCP-1的水平、COX-2、PEG2、TGF-β1、Smad2和Smad3表达水平显著增加(P<0.05)。与模型组相比,低剂量组、高剂量组的腺瘤发生数量、结直肠组织的IL-6 、TNF-α和MCP-1的水平、COX-2、PEG2、TGF-β1、Smad2和Smad3表达水平显著降低,抑瘤率显著增加(P<0.05),并呈剂量依赖性。与高剂量组相比,激动剂组的腺瘤发生数量、结直肠组织的IL-6 、TNF-α和MCP-1的水平、COX-2、PEG2、TGF-β1、Smad2和Smad3表达水平显著增加,抑瘤率显著降低(P<0.05)。结论:补阳还五汤能抑制炎症介质COX-2和PGE2,以及炎症因子IL-6、TNF-α和MCP-1的水平,进而抑制ApcMin/+小鼠CRA的生长,其机制可能是通过抑制TGF-β1/Smad信号通路实现。

      Objective: The aim of this study was to investigate the inhibitory effect of tonifying Yang Returning Five Soup on the growth of colorectal adenoma (CRA) in mice by constructing a mouse model of colorectal adenoma and to deeply analyze its relationship with the transforming growth factor-β1 (TGF-β1)/Smad signaling pathway. METHODS: Ten wild-type C57BL/6J mice were used as the normal group, and 40 Apcmin/+ mice were divided into four groups of 10 mice each according to the randomized numerical table method: the model group, the low-dose group, the high-dose group and the agonist group. The number of colorectal adenomas in each group of mice was observed and recorded.HE staining was performed to observe the pathological changes in the colorectum. Detect the content of IL-6, TNF-α and MCP-1 in colorectal tissues. Immunohistochemistry detected COX-2 and PEG2 expression in colorectal tissues. Protein immunoblotting was employed to detect the expression levels of proteins associated with the TGF-β1/Smad signaling pathway.. RESULTS: Compared with the normal group, a large number of adenomas were observed in the colorectal tissue of the model group. The levels of IL-6, TNF-α and MCP-1, and the expression levels of COX-2, PEG2, TGF-β1, Smad2 and Smad3 in the colorectal tissue of the model group were significantly increased (P<0.05). When compared to the model group, the low-dose group and the high-dose group exhibited a significant reduction in the number of adenomas, the levels of IL-6, TNF-α, and MCP-1 in colorectal tissues, as well as the expression levels of COX-2, PEG2, TGF-β1, Smad2, and Smad3. Additionally, the tumor inhibition rate in these two groups was significantly elevated (P<0.05) in a dose-dependent manner. Compared with the high-dose group, the number of adenomas, the levels of IL-6, TNF-α and MCP-1 in colorectal tissues, the expression levels of COX-2, PEG2, TGF-β1, Smad2 and Smad3 in the agonist group were significantly increased, and the tumor inhibition rate was significantly decreased (P<0.05). CONCLUSION: Tonic Yang Returning Five Soup inhibited the levels of inflammatory mediators COX-2 and PGE2, as well as inflammatory factors IL-6, TNF-α and MCP-1, and thus inhibited the growth of CRA in ApcMin/+ mice, which was probably achieved by the mechanism of inhibiting TGF-β1/Smad signaling pathway.
