      目的利用两样本孟德尔随机化方法探究遗传水平上咖啡摄入与食管癌发病风险之间的因果关系。方法该研究于 2023年 9—11月从开放全基因组关联研究数据库获取咖啡摄入和食管癌相关的单核苷酸多态性( SNP)作为工具变量并进行筛选,采用逆方差加权法( IVW)、加权中位数( WM)、 MR-Egger等方法评估其因果关联,并进行敏感性分析。结果 IVW发现咖啡摄入和食管癌发病风险之间存在关联性[OR=4.430,95%CI:(1.639,11.978),P=0.003]。敏感性分析表明结果具有稳健性。全部 SNP的 F值大于 10,提示不存在弱工具变量偏倚。结论咖啡摄入会增加欧洲人群食管癌患病风险。
英文摘要:
      Objective To investigate the genetic causal relationship between coffee intake and the risk of esophageal cancer using two-sample Mendelian randomization.Methods In this study, single nucleotide polymorphisms (SNPs) associated with coffee intakeand esophageal cancer were obtained from an open genome-wide association studies (GWAS) database between September and Novem-ber 2023 and subjected to screening. The inverse variance weighted (IVW) method, weighted median (WM) approach, and MR-Egger regression were employed to evaluate the causal relationship, followed by sensitivity analyses.Results The IVW method revealed a significant association between coffee intake and the risk of esophageal cancer [OR=4.430, 95%CI: (1.639, 11.978), P=0.003]. Sensitiv-ity analysis indicated that the results were robust. The F-statistics for all SNPs exceeded 10, suggesting the absence of weak instrument bias.Conclusion Coffee intake increases the risk of esophageal cancer in European population.
