目的探究环境温度变化对特应性皮炎(AD)小鼠炎症状态的影响。方法研究时间 2023年 11月至 2024年 6月, 24只雌性昆明鼠采用随机数字表法分为四组,包括对照组和 AD组。 AD组连续 7d涂布卡泊三醇( MC903),对照组涂布等剂量无水乙醇,第 7天、第 14天时分别收集相关样品,对照组置于 24 ℃暴露仓, AD组分别置于 24 ℃、10 ℃及 40 ℃暴露仓中,共计 2周。取小鼠耳部组织进行苏木精 -伊红( HE)染色。采用酶联免疫吸附分析( ELISA)检测小鼠血清中胸腺基;淋巴细胞生成素(TSLP)、白细胞介素( IL)-4、IL-6的表达情况;蛋白质印迹法检测组织中高迁移率族蛋白 1(HMGB1)和晚期糖基化终产物受体(RAGE)蛋白表达水平。结果通过 MC903构建小鼠 AD模型,与对照组相比,经不同温度暴露后,小鼠耳部皮肤红肿加重,炎症细胞浸润明显; AD组血清 IL-4、IL-6、TSLP含量升高, IL-6、TSLP含量随温度的升高或降低呈现升高趋势;而不同温度之间,高温组 IL-6[15.52(13.62,23.71)ng/L比 11.91(11.33,12.43)ng/L]、 TSLP含量[1 579.82(1 391.71,1 868.53)mg/L比 326.19(235.80,420.01)mg/L]均高于常温组,差异有统计学意义( P<0.05);低温或高温环境下小鼠组织中 HMGB1、RAGE表达均呈现升高趋势,差异有统计学意义( P<0.05)。结论高温和低温条件下, AD小鼠 IL-4、IL-6、TSLP、HMGB1、RAGE表达均呈现升高趋势,提示极端温度变化可能通过 HMGB1/RAGE信号通路影响 AD的发病。 |

Objective To explore the effect of ambient temperature change on the inflammatory state of atopic dermatitis mice.Meth. ods The research period was from November 2023 to June 2024,24 female Kunming rats were randomly divided into 4 groups, includ-ing control group and atopic dermatitis model group.AD group was coated with calcipotriol (MC903) continuously for 7 days, and thecontrol group was coated with equal dose of anhydric ethanol. Relevant samples were collected on the 7th day, and then the controlgroup was exposed to 24 ℃ temperature exposure chamber, Atopic dermatitis AD groups were exposed to 24 ℃, 10 ℃ and 40 ℃ respec-tively for a total of 2 weeks. The ear tissue of mice was stained with hematoxylin-eosin. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression of TSLP, interleukin (IL)-4 and IL-6 in serum of mice. Western blotting was used to detect the proteinexpression levels of high mobility group protein 1 (HMGB1) and receptor for advanced glycation end products (RAGE) in tissues.Re. sults The mice model of atopic dermatitis was established by MC903. Compared with the control group, the ear skin was more red andswollen, and the inflammatory cell infiltration was obvious after exposure to different temperatures. The contents of IL-4, IL-6 and TSLP in serum of AD group were basically increased, and the contents of IL-6 and TSLP showed an increasing trend with the increase or de-crease of temperature.The contents of IL-6[15.52(13.62,23.71)ng/L vs. 11.91(11.33,12.43)ng/L] and TSLP[1 579.82(1 391.71,1 868.53) mg/L vs. 326.19(235.80,420.01)mg/L] in high temperature group were higher than those in normal temperature group, and the differ-ence was statistically significant(P<0.05). The expressions of HMGB1 and RAGE in mice tissues at low temperature or high tempera-ture showed an increasing trend, and the differences were statistically significant(P<0.05).Conclusion The expressions of IL-4, IL-6,TSLP, HMGB1 and RAGE in AD mice increased under both high and low temperature conditions, suggesting that extreme temperaturechanges may affect the pathogenesis of AD through the HMGB1/RAGE signaling pathway. |
