| N6-甲基腺苷(N6-methyladenosine, m6A)修饰作为真核生物mRNA中最普遍的表观转录组修饰,通过其调控因子(书写器、擦除器、阅读器)精密调控基因表达,在肿瘤发生发展中起关键作用。本综述系统阐述了m6A修饰在食管癌(Esophageal Carcinoma, EC)中的多重作用:它不仅驱动肿瘤细胞的增殖、侵袭、转移及代谢重编程,还通过塑造免疫抑制微环境与介导免疫逃逸促进肿瘤进展。此外,m6A通过调控铁死亡等过程影响EC的化疗耐药与放疗反应。最后,我们探讨了m6A调控因子高选择性抑制剂的开发、在逆转肿瘤细胞耐药性及联合治疗中的巨大潜力,为EC的预后判断和精准治疗提供新思路。 |

| N6-methyladenosine (m6A) modification, as the most prevalent epigenetic modification of mRNA in eukaryotes, precisely regulates gene expression through its regulatory factors (writers, erasers, and readers), playing a pivotal role in tumorigenesis and progression. This review systematically elucidates the multifaceted roles of m6A modification in esophageal carcinoma (EC): it not only drives tumor cell proliferation, invasion, metastasis, and metabolic reprogramming, but also promotes tumor progression by shaping an immunosuppressive microenvironment and mediating immune evasion. Furthermore, m6A influences EC chemotherapy resistance and radiotherapy response by regulating processes such as ferroptosis. Finally, we explore the development of highly selective inhibitors targeting m6A regulators and their significant potential in reversing tumor cell resistance and enhancing combination therapies, offering novel insights for prognostic assessment and precision treatment in EC. |
