      目的探讨茯苓酸对哮喘小鼠气道黏液分泌、气道炎症和细胞外信号调节蛋白激酶( ERK)/核因子 κB(NF-κB)通路的影响。方法 2023年 2—11月, 40只 C57小鼠分为 PBS组、 Asthma组、茯苓酸组及 Asthma+茯苓酸组,应用卵清蛋白建模。苏木精 -伊红( HE)及过碘酸希夫( PAS)染色观察小鼠肺组织病理变化;流式细胞术检测肺组织各类免疫细胞数量及比例;酶联免疫分析检测肺组织白细胞介素( IL)-4、IL-13含量;蛋白质印迹法检测肺组织磷酸化 NF-κB(p-NF-κB)、 NF-κB、磷酸化 ERK1/2(p-ERK1/2)、 ERK1/2蛋白表达并计算比值。结果 PBS组、 Asthma组、茯苓酸组及 Asthma+茯苓酸组 p-NF-κB/NF-κB水平依次为 0.93±0.06、1.33±0.07、0.71±0.03、0.64±0.03,p-ERK/p-ERK水平依次为 0.60±0.04、1.05±0.01、0.88±0.03、0.85±0.11;在卵清蛋白诱导的哮喘小鼠中,茯苓酸可以改善肺组织炎症,降低炎症指数,减轻气道黏液分泌,降低炎症因子 IL-4及 IL-13含量,减小 p-NF-κB/NF-κB、p-ERK/ERK(P<0.05)。结论茯苓酸可减轻哮喘小鼠气道炎症,机制与抑制 ERK/NF-κB信号通路,减轻气道黏液分泌有关。

      Objective To explore the effects of pachymic acid (PA) on airway mucus secretion, airway inflammation and extracellular signal-regulated kinase (ERK) / nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) signaling pathway in asthmatic mice.Methods From February 2023 to November 2023, forty C57 female mice were randomly assigned to blank control group (PBS),asthma model group (Asthma), PA drug control group (PA), and PA experimental group (Asthma + PA). The animal model of allergic asth-ma was constructed by applying ovalbumin. Hematoxylin-eosin (HE) and Periodic acid Schiff (PAS) staining was used to observe the path-ological changes in the lung tissue of mice. Flow cytometry analysis was used to measure the frequency and number of immune cell popu-lations in lung tissue. Enzyme-linked immunosorbent assay (ELISA) was used to determine the contents of interleukin-4 (IL-4) and IL-13 in lung tissue. Western blotting method was used to detect the protein expression levels of phosphorylated NF-κB (p-NF-κB), NF-κB, p-ERK1/2, and ERK1/2 in lung tissue and to calculate their relative ratios. Results The ratios of p-NF-κB/NF-κB and p-ERK/ERK inPBS, Asthma, PA, and Asthma + PA groups were 0.93±0.06, 1.33±0.07, 0.71±0.03, 0.64±0.03 and 0.60±0.04, 1.05±0.01, 0.88±0.03,0.85±0.11, respectively. In ovalbumin-induced asthmatic mice, pachymic acid alleviated lung tissue inflammation, reduced the inflamma-tory index, attenuated airway mucus secretion, lowered the levels of inflammatory factors IL-4 and IL-13, and diminished the p-NF-κB/ NF-κB and p-ERK/ERK ratios (P < 0.05).Conclusion Pachymic acid can reduce airway inflammation in asthmatic mice, whose mecha-nism may be related to the inhibition of ERK/ NF-κB signaling pathway activation and attenuation of airway mucus secretion.
