      Notch信号通路是一种高度保守的细胞信号转导途径,在胚胎发育、细胞分化、增殖以及凋亡过程中发挥重要作用。近年来, Notch信号通路在肝癌发生发展中的作用备受关注。大部分数据证明了 Notch信号通路的促癌作用,但在调控巨噬细胞分化与 T细胞毒性方面 Notch又发挥了抗肿瘤作用。此外, Notch信号通路与其他信号通路(如 Wnt和 PI3K/Akt等)在肝癌的发生发展中存在复杂的交互作用,对肝癌的侵袭、转移及耐药性具有重要影响。针对 Notch信号通路的治疗策略,包括通路抑制剂、单克隆抗体及小分子化合物等,已在多种前临床及临床研究中展现出潜力。 Notch信号通路的双重作用特点及其多层次调控机制给临床转化带来了挑战。该文综述了 Notch信号通路在肝癌发生发展中的作用机制、与其他信号通路的交互关系及其作为治疗靶点的最新研究进展,以期为 Notch靶向治疗的开发及肝癌治疗策略的优化提供参考。
英文摘要:
      The Notch pathway is a conserved pathway that plays an important role in embryonic development, cell differentiation, pro-liferation and apoptosis. The role of the Notch pathway in liver cancer is a growing area of interest.Most data show that the pathway hasa pro-cancer effect, but it also plays an anti-tumor role in regulating macrophage differentiation and T-cell toxicity. In addition, theNotch signaling pathway also interacts with other pathways (including Wnt and PI3K/Akt) in the development of hepatocellular carcino-ma, affecting invasion, metastasis and drug resistance. Treatments targeting the Notch pathway, including drugs, antibodies, and smallmolecule compounds, have demonstrated promise in studies. However, the dual-action feature of Notch signaling pathway and its multi-level regulatory mechanism pose a challenge for clinical translation. In this article, we review the Notch signaling pathway's role in liv-er cancer development, its interactions with other pathways, and recent advances as a therapeutic target, aiming to support Notch-target-ed therapy and optimize liver cancer treatment strategies.
