| 目的探讨环状 RNA淀粉样前体样蛋白 2(circAPLP2)通过靶向微 RNA(miR)-455-3p对子宫内膜癌( EC)恶性生物学行为的作用。方法 2023年 9月至 2024年 1月,从 TCGA-UCEC中下载 miRNA表达数据,筛选目标 miRNA。Starbase数据库预测 miRNA上调调控分析。利用实时荧光定量逆转录聚合酶链式反应( qRT-PCR)检测 miR-455-3p和 circAPLP2 mRNA的表达。蛋白质印迹法分析细胞周期相关蛋白和 Notch信号通路相关蛋白的表达。利用生信预测以及双萤光素酶报告基因分析以验证 miR-455-3p与 circAPLP2的靶向关系。克隆形成实验检测 EC细胞的增殖,流式细胞术检测 EC细胞周期进程,划痕愈合实验测定 EC细胞的迁移, Transwell分析法评估 EC细胞的侵袭能力。结果 miR-455-3p在 EC细胞中低表达( 0.62±0.08、0.51±0.06、0.71±0.09、0.46±0.06比 1.01±0.11)明显低于人正常子宫内膜上皮细胞系的 1.01±0.11。过表达 miR-455-3p抑制 EC细胞的增殖、迁移和侵袭,敲低 miR-455-3p的表,达则增强 EC细胞的增殖、迁移和侵袭。此外, circAPLP2作为 miR-455-3p的分子海绵。敲低 circAPLP2和 PBK则能逆转过表达 miR-455-3p对 EC细胞增殖和转移的抑制作用。结论 circAPLP2通过靶向 miR-455-3p激活 Notch信号通路促进 EC的增殖和转移,这可能为认识 EC恶性进展提供了一个新的途径。 |

| Objective To investigate the effect of circAPLP2 on the malignant biological behavior of endometrial carcinoma (EC) by targeting miR-455-3p.Methods The expression of miRNA was downloaded from TCGA-UCEC to screen target miRNAs from Septem-ber 2013 to January 2014. The up-regulation analysis of miRNA was predicted by Starbase database. The expression of miR-455-3p and circAPLP2 mRNA was detected by quantitative reverse transcription polymerase chain reaction (qRT-PCR). The expression of cellcycle related proteins and Notch signaling pathway related proteins was analyzed by Western blotting. The targeting relationship be-tween miR-455-3p and circAPLP2 was verified by bioinformatics prediction and dual luciferase reporter gene analysis. The prolifera-tion of EC cells was detected by colony formation assay, the cell cycle progression was detected by flow cytometry, the migration of ECcells was determined by scratch healing assay, and the invasion ability of EC cells was evaluated by Transwell assay.Results MiR-455-3p was lowly expressed in EC cells (0.62±0.08,0.51±0.06,0.71±0.09,0.46±0.06 vs. 1.01±0.11). Overexpression of miR-455-3p in-hibited the proliferation, migration and invasion of EC cells, while knockdown of miR-455-3p enhanced the proliferation, migration and invasion of EC cells. In addition, circAPLP2 served as a molecular sponge for miR-455-3p. Knockdown of circAPLP2 and PBK re-versed the inhibitory effect of overexpression of miR-455-3p on the proliferation and metastasis of EC cells.Conclusion CircAPLP2 promotes the proliferation and metastasis of EC by targeting miR-455-3p to activate Notch signaling pathway, which may provide a new way to understand the malignant progression of EC. |
