| 急性肺损伤( ALI)与急性呼吸窘迫综合征( ARDS)是由多种直接或间接因素损害所导致的一种严重的炎症性疾病,者的共同病理特征为顽固性低氧血症、肺顺应性下降、肺血管通透性增加及通气 /血流比例失调。 ALI/ARDS在临床上病死率两高,目前的治疗策略以对症支持治疗为主,尚未出现一种有效针对 ALI/ARDS的治疗手段。组蛋白去乙酰化酶( HDACs)是一类蛋白酶,其通过对染色体结构的重塑来调控基因的表达,广泛参与细胞的生理发育过程。 HDAC抑制剂( HDACi)已被广泛应用于癌症、炎症及免疫系统疾病中。同时,越来越多的证据表明靶向 HDACs是治疗 ALI/ARDS的潜在途径。现阐述 HDACs在 ALI/ARDS中作用的研究进展,为临床靶向治疗 ALI/ARDS提供理论依据。 |

| Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) are serious inflammatory diseases caused by a vari.ety of direct or indirect factors, and their common pathological features include intractable hypoxemia, decreased lung compliance, in.creased pulmonary vascular permeability and dysregulation of ventilation/blood flow ratio. The clinical mortality of ALI/ARDS is high,and the current treatment is mainly symptomatic supportive treatment, and there is not yet an effective treatment for ALI/ARDS. His.tone deacetylase (HDACs) is a kind of protease, which regulates gene expression by remodeling chromosome structure and widely par.ticipates in the physiological development of cells. HDAC inhibitors (HDACi) are widely participated in the treatment of cancer, inflam.matory and immune diseases, there is growing evidence that targeting HDACs is a potential pathway for ALI/ARDS. In this review, theresearch progress on the role of histone deacetylase in ALI/ARDS will be reviewed to provide a theoretical basis for clinical targetedtreatment of ALI/ARDS. |
