      目的探讨一种候选新药对皮肤光损伤模型大鼠的防治作用。方法 2022年 5—11月取 42只雄性大鼠采用抽签法随机分为正常对照组、模型对照组、自然愈合组、候选新药预防组、阳性药物预防组、候选新药治疗组、阳性药治疗组。根据不同组别对大鼠进行不同方案紫外线照射及候选新药、阳性药处理,观察皮肤破损变化、病理切片变化,采用酶联免疫试剂盒测定大鼠表皮超氧化物歧化酶、羟脯氨酸、过氧化氢酶等生化指标的变化。结果照射的大鼠皮肤外观及组织结构与空白组相比可见显著改变;给药后的大鼠皮肤损伤修复优于未给药组皮肤;皮肤光老化过程中大鼠体内羟脯氨酸( hydroxyproline,HYP)、超氧化物歧化酶( superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)等指标随皮肤损伤进展发生相应变化,其中 HYP浓度在阳性药治疗组( 1.02±0.15)mg/L、候选新药治疗组( 1.04±0.21)mg/L相较自然愈合组( 1.24±0.03)mg/L更低( P<0.05)SOD浓度在阳性药治疗组( 284.86±72.97)ng/L、候选新药治疗组( 295.33±99.93)ng/L较自然愈合组( 71.24±117.43)ng/L浓度有所升,高( P<

      Objective To investigate the protective and therapeutic effects of a novel drug candidate on skin photodamage using a rat model.Methods From May to November 2022, forty-two male rats were randomly assigned to one of seven groups: the normal controlgroup, the model control group, the natural healing group, the candidate new drug prophylaxis group, the positive drug prophylaxisgroup, the candidate new drug treatment group, and the positive drug treatment group. Following protocol-specific ultraviolet irradiationexposure, the respective treatments were administered. Skin damage was assessed through visual inspection and histopathological anal-ysis. Additionally, an enzyme-linked immunosorbent assay (ELISA) was utilized to measure changes in key biochemical markers suchas superoxide dismutase (SOD), hydroxyproline (HYP), and catalase (CAT) levels in the epidermis.Results Compared with the blankgroup, significant changes were observed in the skin appearance and tissue structure of rats in the irradiated group. The skin damage re-pair in the drug-administered group was superior to that in the non-drug-administered group. During the process of skin photoaging inrats, indicators such as HYP, SOD, and CAT in the body changed correspondingly with the progression of skin damage. Specifically, theHYP concentration in the positive drug treatment group (1.02±0.15) mg/L and the candidate new drug treatment group (1.04±0.21) mg/L was lower than that in the spontaneous healing group (1.24±0.03) mg/L (P<0.05). The SOD concentration in the positive drug treat-ment group (284.86±72.97) ng/L and the candidate new drug treatment group (295.33±99.93) ng/L was higher than that in the spontane-ous healing group (71.24±117.43) ng/L (P<0.05). These results indicated that the candidate new drug compound activated the body'ssuperoxide clearance process to achieve therapeutic effects, demonstrating efficacy in preventing and treating skin photo damage com-parable to that of MEBO (moist exposed burn ointment).Conclusion Our findings indicate that the new drug compound, developedthrough molecular simulation and virtual screening techniques, exhibits promising protective and therapeutic effects against skin photo.aging.
