      目的探究向天果提取物对白色脂肪细胞棕色化的影响及机制。方法该研究起止时间为 2023年 6月至 2024年 2月。首先通过细胞计数试剂盒(CCK-8)法检测细胞活力,以确定向天果提取物的安全浓度与处理时间;随后采用蛋白质印迹法和实时荧光定量逆转录聚合酶链反应(qRT-PCR)分别检测棕色化及自噬相关蛋白与 mRNA的表达水平;通过 Bodipy与油红 O染色观察脂滴积累情况,并利用线粒体染料检测线粒体丰度;采用 MDC染色评估自噬小体的形成。最后,通过加入自噬激活剂雷帕霉素进行实验,研究证实向天果提取物促进白色脂肪细胞棕色化的作用机制。结果对照组、 50 g/L向天果提取物组 PRDM16蛋白表达量分别为 1.00±0.07和 1.64±0.13,PGC-1α蛋白表达量分别为 1.00±0.10和 2.55±0.17;UCP-1蛋白表达量分别为 1.00±0.11和1.81±0.16,表明向天果提取物能够促进棕色化相关蛋白( P<0.05)对照组、 50 g/L向天果提取物组油红 O染色后 OD510值分别为 1.00±0.07和 0.47±0.05,表明抑制脂滴积累,对照组、 50 g/L向果提天取物组 Beclin-1蛋白表达量分别为 1.00±0.09和 0.25±0.05,p62蛋白表达量分别为 1.00±0.07和 1.96±0.11;表明向天果提取物能够抑制自噬水平,同时,向天果提取物能够促进棕色化与自噬相关 mRNA表达,增加线粒体丰度,抑制自噬小体的表达,最后,自噬激活剂逆转向天果提取物对脂肪细胞自噬抑制的同时,促棕色化作用也发生了逆转。结论向天果提取物能够通过抑制自噬促进白色脂肪细胞棕色化。

      Objective To explore the effect and mechanism of Swietenia macrophylla extracts on the browning of white adipocytes. Methods The study was conducted from June 2023 to February 2024. Firstly, cell viability was detected by Cell Counting Kit-8 (CCK-8) assay to determine the safe concentration and treatment time of Swietenia macrophylla extracts. Subsequently, Western blot and qRT-PCR were used to detect the expression levels of browning and autophagy-related proteins and mRNA, respectively. The accumulation oflipid droplets was observed by Bodipy and oil red O staining, and the mitochondrial abundance was detected by mitochondrial dye. Theformation of autophagosomes was evaluated by MDC staining. Finally, rescue experiments with the addition of the autophagy activator ra-pamycin confirmed that the effect of Swietenia macrophylla extracts in promoting the browning of white adipocytes was achieved by in-hibiting the autophagy pathway.Results The protein expression levels of PRDM16, PGC-1α and UCP-1 were 1.00±0.07, 1.00±0.10 and 1.00±0.11 in the control group and 1.64±0.13, 2.55±0.17, and 1.81±0.16 in the 50 g/L Swietenia macrophylla extract group, respec-tively, indicating that Swietenia macrophylla extracts could promote the expression of browning-related proteins (P<0.05). The OD510 val-ues after oil red O staining were 1.00±0.07 and 0.47±0.05 in the control group and the 50 g/L Swietenia macrophylla extract group, re-spectively, indicating Swietenia macrophylla extracts inhibited lipid droplet accumulation. The protein expression levels of Beclin-1 and p62 were 1.00±0.09 and 1.00±0.07 in the control group and 0.25±0.05 and 1.96±0.11, respectively, suggesting Swietenia macrophylla extracts could suppress autophagy levels. Additionally, Swietenia macrophylla extracts upregulated the browning-and autophagy-related mRNA expression, increased mitochondrial abundance, and inhibited the expression of autophagosomes. Finally, the autophagy activa-tor reversed both the inhibitory effect of Swietenia macrophylla extracts on adipocyte autophagy and its promoting effect on browning. Conclusion The extracts of Swietenia macrophylla promotes the browning of white adiposes by inhibiting autophagy.
