      目的:探讨桦褐孔菌多糖对糖尿病肾病小鼠脂质代谢的影响并初步探讨其分子机制。方法:将36只雄性db/db小鼠随机分为模型组、AKT信号通路抑制剂MK-2206组、桦褐孔菌多糖组、桦褐孔菌多糖+AKT信号通路激动剂SC79组,每组9只。另选取9只db/m雄性小鼠为对照组。检测各组小鼠的体重、空腹血糖(FBG),检测血清中血尿素氮(BUN)、血肌酐(Scr)、尿蛋白、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平,HE染色及PAS染色检测肾组织的病理学损伤情况,油红O染色检测肾组织的脂滴沉积情况,Western blot检测肾组织中PINK1、Parkin、LC3、Beclin1、p62、FOXO1蛋白的表达及p-AKT/AKT、p-p38 MAPK/p38 MAPK的比值。结果:相对于对照组,模型组小鼠的体质量、FBG、BUN、Scr、尿蛋白、TC、TG、LDL-C、p62、p-AKT/AKT比值、p-p38 MAPK/p38 MAPK比值升高(P<0.05),HDL-C、PINK1、Parkin、Beclin1、LC3II/Ⅰ比值、FOXO1降低(P<0.05),肾组织呈现显著的病理学损伤及脂质沉积;相对于模型组,MK-2206组、桦褐孔菌多糖组小鼠的体质量、FBG、BUN、Scr、尿蛋白、TC、TG、LDL-C、p62、p-AKT/AKT比值、p-p38 MAPK/p38 MAPK比值降低(P<0.05),HDL-C、PINK1、Parkin、Beclin1、LC3II/Ⅰ比值、FOXO1升高(P<0.05),肾组织显著的病理学损伤及脂质沉积减轻;SC79能在一定程度上逆转桦褐孔菌多糖对糖尿病肾病小鼠的作用(P<0.05)。????????????? 结论:桦褐孔菌多糖能够通过促进线粒体自噬来减轻糖尿病肾病小鼠的脂质代谢紊乱及肾脏组织的脂质沉积,其机制可能与抑制AKT/FOXO1/MAPK信号通路有关。
英文摘要:
      Objective: To explore the effect of Inonotus obliquus polysaccharides on lipid metabolism in diabetic nephropathy mice, and to explore its molecular mechanism. Methods: Thirty-six male db/db mice were randomly divided into model group, AKT signaling pathway inhibitor MK-2206 group, Inonotus obliquus polysaccharides group, and Inonotus obliquus polysaccharides + AKT signaling pathway agonist SC79 group, with 9 mice in each group, and 9 db/m male mice were selected as the control group. Body weight and fasting blood glucose (FBG) of mice in each group were measured. The levels of blood urea nitrogen (BUN), serum creatinine (Scr), urine protein, total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) in serum were detected. HE staining and PAS staining were used to detect the pathological damage of renal tissue. Oil red O staining was used to detect lipid droplet deposition in kidney tissue. Western blot was used to detect the expression of PINK1, Parkin, LC3, Beclin1, p62 and FOXO1 proteins and the ratio of p-AKT/AKT and p-p38 MAPK/p38 MAPK in renal tissue.
