| 目的探讨肝细胞生长因子受体( HGFR)抗体偶联金纳米颗粒对 HeLa细胞的靶向作用及放射治疗的增敏作用。方法收集 2022年 1—12月烟台毓璜顶医院 10例宫颈腺癌及癌旁组织样本。合成金纳米颗粒后用透射电镜对其进行观察并采集图像。对 HeLa细胞进行免疫染色后使用荧光显微镜观察 HGFR定位及表达情况并采集图像。对金纳米颗粒进行聚乙二醇化并偶联 HGFR抗体后通过细胞计数试剂盒( CCK-8)法进行细胞毒性检测。使用 X线对各组细胞进行照射,通过 CCK8法测定细胞抑制率,通过蛋白质印迹法检测凋亡相关蛋白表达情况。结果该研究成功合成了金纳米颗粒,并通过双官能团聚乙二醇对其进行修饰及偶联靶向抗体。金纳米颗粒的半数抑制浓度( IC50)为 3.6 nmol/L,金纳米颗粒组细胞存活率( 60.0±27.5)%,而靶向金纳米颗粒的细胞毒性较前者降低,其 IC50为 8.8 nmol/L,靶向金纳米颗粒组细胞存活率( 73.2±24.5)%。X线照射后,随照射剂量的增加,细胞存活抑制率逐渐升高,在相同照射剂量的条件下,靶向金纳米颗粒组的抑制率均显著高于其他组: X线照射组细胞增殖抑制率( 21.5±13.0)%,金纳米颗粒联合 X线照射组细胞增殖抑制率( 27.7±17.4)%,靶向金纳米颗粒联合 X线照射组细胞增殖抑制率( 43.0±25.2)%。靶向金纳米颗粒联合放射治疗组中凋亡相关蛋白表达量均高于其他两组。结论 HGFR抗体偶联金纳米颗粒对 HeLa细胞的放射治疗具有一定的增敏作用。 |

| Objective To investigate the targeting effect of hepatocyte growth factor receptor (HGFR) antibody coupled with goldnanoparticles on HeLa cells and the sensitizing effect on radiotherapy.Methods Ten cervical adenocarcinoma and adjacent normaltissue samples from Yantai Yuhuangding Hospital were collected between January and December 2022. The synthesized gold nanopar-ticles were observed by transmission electron microscopy and the images were collected. After immunostaining, the localization and ex-pression of HGFR in HeLa cells were observed by fluorescence microscope and the images were collected. The gold nanoparticles werePEGylated and coupled with HGFR antibody for cytotoxicity detection by CCK8 method. X-rays were used to irradiate the cells in eachgroup, and the cell inhibition rate was determined by CCK8 assay, and the expression of apoptosis-related proteins was detected by pro-tein blotting (western blot).Results In this study, gold nanoparticles were successfully synthesized, and subsequently modified by bi-functional PEG, followed by coupling to targeting antibodies. The half inhibitory concentration (IC50) of gold nanoparticles was 3.6 nmol/L, and the cell survival rate of gold nanoparticle group was (60.0±27.5)%, while the cytotoxicity of targeted gold nanoparticles was re-duced compared with the former, the IC50 was 8.8 nmol/L, and the cell survival rate of the targeted gold nanoparticle group was (73.2± 24.5)%. After X-ray irradiation, the inhibition rate of cell survival increased gradually with the increase of irradiation dose, and the inhi-bition rate of the targeted gold nanoparticle group was significantly higher than that of the other groups in all of the groups under thesame irradiation dose conditions. The cell proliferation inhibition rate was (21.5±13.0)% in the X-ray irradiation group, (27.7±17.4)% in the gold nanoparticles plus X-ray irradiation group, and (43.0±25.2)% in the targeted gold nanoparticles plus X-ray irradiation group. The expression of apoptosis-related proteins in the targeted gold nanoparticles plus radiotherapy group was higher than that in the other two groups. Conclusion HGFR antibody-coupled gold nanoparticles sensitize HeLa cells for radiation therapy. |
