| 【】Tumor metabolic reprogramming and immune evasion are central characteristics of malignant tumor development. Adenosine triphosphate citrate lyase (ATP citrate lyase, ACLY), serving as a critical link between carbohydrate metabolism and epigenetic regulation, exhibits abnormally high expression in various solid tumors. Recent studies have demonstrated that ACLY not only meets the metabolic demands of tumor cells by providing acetyl-CoA but also regulates the expression of immune checkpoint genes through histone acetylation modifications, thereby mediating tumor immune evasion.This review aims to summarize the pivotal role of ACLY in regulating the tumor immune microenvironment, investigate the molecular mechanisms underlying the synergistic effect of ACLY-targeting therapies combined with immune checkpoint inhibitors in overcoming drug resistance, and outline future challenges and strategies for clinical translation. |
