      目的 比较长沙地区不同基原蒲黄(水烛香蒲与长苞香蒲)花粉中化学成分及含量的差异,为其质量控制体系的提升及临床精准用药提供科学依据。方法 采用扫描电镜观察2种蒲黄花粉显微形态;采用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF-MS)对2种蒲黄花粉化学成分进行定性鉴定;建立超高效液相色谱-三重四极杆质谱(UPLC-QQQ-MS)多反应监测(MRM)方法,对香蒲新苷和异鼠李素进行定量测定。结果 定性分析共鉴定出27种化学成分,包括14种黄酮类、6种有机酸类和7种其他类成分;两种基原蒲黄共有成分25种。方法学考察显示,香蒲新苷和异鼠李素线性关系良好(R²>0.99)。长沙松雅湖长苞香蒲中香蒲新苷和异鼠李素含量总和为13562.215 ng/mL,长沙西湖水烛香蒲为7645.458 ng/mL。两者在两种化合物及化合物总和上均表现出显著性差异(p<0.001)。结论 长沙西湖水烛香蒲与长沙松雅湖长苞香蒲在化学成分种类上高度相似,但长苞香蒲黄酮类指标成分积累水平更高。本研究可为蒲黄多基原药材质量评价与分选应用提供参考。

      Objective To compare the differences in chemical constituents in pollen of Typha angustifolia L. and Typha domingensis Pers. (two original sources of Puhuang) in Changsha, so as to provide a scientific basis for improving the quality control system and precise clinical medication of Puhuang. Methods Scanning electron microscopy was used to observe pollen micro-morphology. UPLC-Q-TOF-MS was used for qualitative identification of chemical constituents, and a UPLC-QQQ-MS/MRM method was established for quantitative determination of typhaneoside and isorhamnetin. Results A total of 27 constituents were identified, including 14 flavonoids, 6 organic acids and 7 other compounds. Twenty-five constituents were shared by the two original species. The method showed good linearity for typhaneoside and isorhamnetin (R²>0.99). The total content of typhaneoside and isorhamnetin was 13562.215 ng/mL in Typha domingensis from Songya Lake, Changsha, and 7645.458 ng/mL in Typha angustifolia from West Lake, Changsha. Both showed significant differences in the two compounds and the sum of the compounds (p < 0.001). Conclusion The two original species are highly similar in constituent categories, while Typha domingensis shows higher accumulation of the selected flavonoid markers. The study provides a reference for quality evaluation and source discrimination of Typhae Pollen.
