      目的 探究T-box转录因子TBX2与鼠双微体基因MDM2在正常增生期内膜、各型增生内膜和子宫内膜样腺癌中的表达及相关性。方法 组织芯片技术结合免疫组化SP法检测92例子宫内膜样腺癌、30例复杂伴不典型性增生内膜、30例单纯性增生内膜及30例正常增生期内膜中TBX2与MDM2蛋白表达。 结果 TBX2阳性表达率在子宫内膜样腺癌(76.1%,70/92) 中显著高于正常增生期内膜及单纯性增生内膜(30.0%,9/30;36.7%,11/30),亦显著高于复杂伴不典型增生内膜(43.3%,13/30),差异均具有统计学意义(P<0.01),TBX2表达过高与病理学分级、肌层浸润及临床分期均有关(P<0.01, P<0.01, P<0.05),与淋巴结转移不具相关性(P>0.05);MDM2蛋白在子宫内膜样腺癌及复杂伴不典型增生中阳性率分别为81.5%(75/92)和43.3%(13/30),显著高于正常子宫内膜及单纯性增生内膜(10.0%,3/30;20.0%,6/30),差异均有统计学意义(P<0.01),MDM2阳性表达与病理学分级、肌层浸润无关,与临床分期及淋巴结转移也不具相关性(P均>0.05)。TBX2与MDM2蛋白表达呈正向相关(rs=0.346,P<0.01)。结论 TBX2和MDM2蛋白在子宫内膜样腺癌中异常高表达,并在其发生及恶化进展中扮演一定角色,为子宫内膜样腺癌的靶点探寻及预后评判提供了新的方向。
英文摘要:
      Objective : To investigate the relationship of the expression of TBX2 and MDM2 proteins in the development of endometrial adenocarcinoma. Methods The expression of TBX2 and MDM2 protein in 92 specimens of endometrial adenocarcinoma, 30 specimens of endometrial complex hyperplasia with atypical hyperplasia, 30 specimens of endometrial simple hyperplasia and 30 specimens of endometrium in normal proliferative phase were detected by immunohistochemistry (SP method) on the tissue microarray. Results There was significant difference in the positive rate of TBX2 protein between endometrial adenocarcinoma and endometrial normal proliferative phase as well as endometrial simple hyperplasia or endometrial complex hyperplasia ( by atypical )(P<0.01),The positive expression of TBX2 was associated with the histological grade(P<0.01),FIGO stage (P<0.05),depth of invasion(P<0.01), and not related to the lymph node metastasis(P>0.05).The positive rate of MDM2 protein in endometrial adenocarcinoma and complex or by atypical hyperplasia was higher than that in endometrium in normal proliferative phase and simple hyperplasia (P<0.01), while the expression of MDM2was independent of the histological grade and depth of invasion, not related to FIGO stage and lymph node metastasis yet (P>0.05).The expression of TBX2 and MDM2 protein were positively correlated in endometrial adenocarcinoma (r=0.346,P<0.01). Conclusion The abnormal expressions of TBX2 and MDM2 might be connected with the development and malignant degree of endometrial adenocarcinoma. The results suggest that TBX2 and MDM2 may be important biomarkers in the detection ofbiological behavior and prognosis in endometrial adenocarcinoma.
