      目的探讨 β1,3-N乙酰氨基葡萄糖基转移酶 8(β3GnT8)表达与胃癌病人新辅助化疗疗效相关性研究。方法选取 2013年 1月至 2014年 7月在武汉市第一医院确诊为原发性胃癌的病人 80例,免疫组织化学法检测胃癌及癌旁组织中 β3GnT8和基质金属蛋白酶 2(MMP-2)的表达,并观察其新辅助化疗疗效和生存状况。采用 χ2检验、 Kaplan-Meier并 log-rank检验及 Cox回归分析数据。结果 β3GnT8和 MMP-2在癌组织中的阳性表达率均高于癌旁组织。 β3GnT8和 MMP-2在低分化、 Ⅲ期、淋巴结转移病人中阳性表达率增高( P<0.05)。 χ2检验结果显示: β3GnT8高表达组新辅助化疗有效率为 59.70%(40/67),且明显低于 β3GnT8低表达组 92.31%(12/13)(χ2=5.088,P=0.024)。 MMP-2高表达组新辅助化疗有效率为 58.46%(38/65),且明显低于 MMP-2低表达组 93.33%(14/15)(χ2=6.515,P=0.011)。多因素 Cox回归分析显示:较低分化程度、 Ⅲ期、淋巴结转移、 β3GnT8阳性表达、 MMP-2阳性表达可增加新辅助化疗胃癌病人死亡风险[HR(95%CI): 1.27(1.24~1.31)1.49(1.29~1.72),1.32(1.15~

      Objective To investigate the association between the expression of β1, 3-N-acetylglucosminyltransferase 8 (β3GnT8) and the efficacy of neoadjuvant chemotherapy in patients with gastric cancer. Methods Eighty patients with primary gastric cancerhospitalized in Wuhan No.1 Hospital from January 2013 to July 2014 were included. The expression of β3GnT8 and matrix metallopro?teinase-2 (MMP-2) in gastric cancer and adjacent tissues were measured by immunohistochemistry, and the efficacy of neoadjuvant che?motherapy and survival were observed. Chi-square test, Kaplan-Meier with log-rank test, and Cox regression were used to analyze the data. Results The positive expression rates of β3GnT8 and MMP-2 in gastric cancer tissues were higher than those in adjacent tis? sues. The positive expression rates ofβ3GnT8 and MMP-2 increased in patients with low differentiation, Ⅲ stage, and lymph node me? tastasis (P < 0.05). Chi-square test showed that the effective rate of neoadjuvant chemotherapy was 59.70% (40/67) in the high expres?sion group ofβ3GnT8, which was significantly lower than 92.31% (12/13) in the low expression group of β3GnT8 (χ2=5.088, P=0.024). The effective rate of neoadjuvant chemotherapy in MMP-2 high expression group was 58.46% (38/65), which was also significantly low? er than 93.33% (14/15) in MMP-2 low expression group (χ2=6.515, P=0.011). Multivariate Cox regression analysis showed that lowerdifferentiation, Ⅲ stage, lymph node metastasis, positive expression of β3GnT8 and positive expression of MMP-2 increased the risk of death in neoadjuvant chemotherapy patients with gastric cancer [HR (95%CI): 1.27 (1.24-1.31), 1.49 (1.29-1.72), 1.32 (1.15-1.51), 1.28 (1.14-1.44), 1.31 (1.15-1.50)]. Conclusion The expression of β3GnT8 in gastric cancer tissues is an independent factor affecting theprognosis of neoadjuvant chemotherapy. The patients with β3GnT8 positive expression have poor chemotherapeutic effect and in?creased risk of death.
