      目的采用高效液相色谱-质谱-多反应监测(UHPLC-QTRAP-MRM)特征轮廓谱结合化学计量学分析对市售不同产地药用紫草进行质量评价,以期对紫草饮片的质量控制提供参考。方法采用 Shimadzu LC20ADXR高效液相色谱仪联合 QTRAP 4000质谱仪、ESI离子源、MRM负离子电离模式,建立药用紫草的特征轮廓谱。采用 MarkerView软件分析提取数据,并进行主成分分析和主成分判别分析,分析紫草品种、产地因素对药材的影响。结果无监督的 PCA模型不能有效区分三种不同种属的紫草;PCA-DA模型能有效区分三种紫草,不同来源的紫草无论是种内还是种间样品,其化学成分的组成和含量均有明显差异,并发现化合物 β,β-二甲基丙烯酰紫草素、紫草呋喃 A、1-甲氧基乙酰紫草素、丙酰紫草素和紫草呋喃 E为三种紫草潜在的代谢差异性成分。结论该方法快捷、灵敏、可靠,可为紫草药材的产地选择及质量控制提供参考方法。
英文摘要:
      Objective To evaluate the quality of medicinal Zicao from different sources based on UHPLC-QTRAP-MRM characteristic chemical profile and chemometrics analysis, and to provide reference for the quality control of medicinal Zicao.Methods The characteristic chemical profile was obtained from the UHPLC-MS/MS system with an LC20ADXR binary pump and a QTRAP 4000 system with an electrospray ionization (ESI) in the negative ion mode. The scan mode was in multiple reaction ion monitoring (MRM) mode.The peak area data were extracted from the peaks of UHPLC-QTRAP-MRM characteristic chemical profile by MarkerView 1.2.1. Therelative peak area data were subjected to principal component analysis (PCA), and principal component discriminant analysis (PCADA).Results The unsupervised PCA model failed to distinguish the sample from different species effectively, however, the PCA-DA model showed that three species were distinguished well and the composition and content of chemical components in medicinal Zicao from different sources were different obviously. The compounds β, β-dimethylacrylshikonin, shikonofuran A, 1-methoxyacetylshikonin, propionylshikonin, shikonofuran E were the potential metabolic markers for distinguishing these three species.Conclusion The method is quicker, more sensitive and reliable, which can be used as a reference for the selection and quality control of medicinal Zicao.
