      目的研究金雀异黄酮( GEN)对缺氧 /复氧诱导的心肌细胞损伤的影响及可能分子机制。方法本研究的起止时间为 2018年 10月至 2019年 11月。建立心肌细胞缺氧 /复氧( H/R)模型( H/R组),以常规培养的细胞作为对照组;用浓度分别为5 mg/L、10 mg/L、20 mg/L的 GEN处理 H9c2细胞后再进行 H/R处理,记为 GEN低、中、高浓度组;将小干扰 RNA阴性对照( siNC)、转胶蛋白 -2(TAGLN2)小干扰 RNA(si-TAGLN2)转染至 H9c2细胞中再进行 H/R处理,记为 H/R+si-NC组、 H/R+si-TAGLN2

      Objective To investigate the effect of genistein on hypoxia/reoxygenation-induced cardiomyocyte injury and its possible molecular mechanism.Methods This research started from October 2018 and ended in November 2019. Cardiomyocyte hypoxia/reoxygenation (H/R) model was established (H/R group), and conventionally cultured cells were used as the control (Con) group. H9c2 wastreated with genistein in concentrations of 5 mg/L, 10 mg/L, and 20 mg/L before the cells were treated with H/R and recorded as low-,medium-and high-concentration genistein. The small interfering RNA negative control (si-NC) and transgelin 2 (TAGLN2) small interfering RNA (si-TAGLN2) were transfected into H9c2 cells and then treated with H/R as H/R+si-NC group, H/R+si-TAGLN2 group; empty plasmid vector (pcDNA) and TAGLN2 overexpressed plasmid (pcDNA-TAGLN2) were transfected into H9c2 cells, treated with20 μg/mL genistein, and then treated with H/R as H/R+GEN+pcDNA group, H/R+GEN+ pcDNA-TAGLN2 group. The kit was used todetect malondialdehyde (MDA) content and superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) activity, flow cytometrywas used to detect apoptosis, and Western blot method was used to detect transgelin 2 (TAGLN2) protein expression.Results Compared with Con group, the apoptosis rate [(25.36±2.14)% vs. (7.25±0.71)%], MDA content [(11.02±1.17) nmol/L vs. (3.54±0.34) nmol/ L], TAGLN2 expression level [(0.83±0.07) nmol/L vs. (0.39±0.03) nmol/L] of H9c2 cells were increased, and SOD and GSH-Px activities were decreased in H/R group.
