| Objective To investigate the expression of long-chain non-coding RNA (lncRNA) deleted in lymphocytic leukemia 2(DLEU2) in tongue squamous cell carcinoma and its effect on the proliferation and invasion of cancer cells.Methods Cancerous tissue and corresponding paracancerous tissue specimens were collected from 22 confirmed cases of tongue squamous cell carcinoma whounderwent surgery in The Third Hospital of Handan City from May 2018 to July 2019. The expression of DLEU2 in tongue squamouscell carcinoma cells and tissues were detected by real-time fluorescence quantitative PCR. The tongue squamous cell carcinoma cell line CAL-27 cells cultured in vitro were assigned into control group (untransfected), empty vector group (empty vector) and DLEU2group (transfected with DLEU2). The expression level of DLEU2 was detected by real-time fluorescent quantitative PCR, cell proliferation by MTT method, and cell invasion by the Transwell assay. Results Compared with adjacent tissues, the expression level of DLEU2 in cancerous tissues was significantly reduced [(0.30±0.02) vs. (1.00±0.00), P<0.05]. The expression levels of DLEU2 in cells of SCC-15, SCC-4 and CAL-27 were significantly reduced in comparison with DLEU2 level in normal human oral keratinocytes (HOK)[(0.35±0.04), (0.48±0.04), (0.26±0.03) vs. (1.00±0.00); P<0.05]. Compared with the control group, the expression level of DLEU2 in thecells of the DLEU2 group was significantly increased, and the cell proliferation and invasion capacities were significantly reduced (P< 0.05).Conclusions The expression of lncRNA DLEU2 was low expressed in cells and tissues of tongue squamous cell carcinoma. Up-regulation of its expression can inhibit the proliferation and invasion of tongue squamous cell carcinoma CAL-27 cells. |
