      目的探讨鱼藤素对卵巢癌 SKOV细胞增殖和凋亡的影响及可能的作用机制。方法本研究于 2019年 3月至 2020年 3月进行,鱼藤素作用 SKOV细胞后,细胞计数试剂盒 -8(CCK-8)检测细胞增殖,流式细胞仪检测细胞凋亡,蛋白免疫印迹(Western blotting)法分别对细胞中细胞周期蛋白依赖性激酶抑制剂 1(P21)、细胞周期蛋白 D1(CyclinD1)、 B淋巴细胞瘤 -2(Bcl-2)及 Bcl-2相关 X蛋白( Bax)、 Kruppel样因子 15(KLF15)蛋白表达水平进行检测,实时荧光定量 PCR(RT-qPCR)检测 KLF15 mRNA表达水平。转染 KLF15过表达载体上调 SKOV细胞中 KLF15表达后,上述相同方法观察上调 KLF15表达对 SKOV细胞增殖、凋亡及 CyclinD1、P21、Bcl-2和 Bax蛋白表达的影响。结果鱼藤素作用 SKOV3细胞后,细胞活性( 0.49±0.04、0.39±0.03、0.30±0.02)、 CyclinD1(0.53±0.05、0.45±0.05、0.21±0.03)及 Bcl-2(0.40±0.04、0.24±0.02、0.16±0.01)蛋白表达水平降低( P<0.05),细胞凋亡率[(6.63±0.69)%、(12.71±1.06)%、(20.27±2.25)%]、 P21(0.23±0.02、0.46±0.04、0.74±0.03)、 Bax(0.20±0.02、0.36±0.02、0.55±0.05)蛋白表达水平升高, KLF15 mRNA和蛋白表达水平升高( P<0.05)。上调 KLF15表达后, SKOV3细胞活性、 CyclinD1及 Bcl-2蛋白表达水平降低( P<0.05)细胞凋亡率、 P21及 Bax蛋白表达水平升高( P<0.05)。下调 KLF15表达后,鱼藤素作用的 SKOV3细胞活性、 CyclinD1及 Bcl-2蛋白表达水平升高( P<0.05)细胞凋亡率、 P21及 Bax蛋白表达水平降低( P<0.05)。结论鱼藤素可抑制卵巢癌 SKOV3细胞增殖,并诱导 SKOV3细胞凋亡作用机制与上调细胞中 KLF15表达有关。

      Objective To investigate the effect of deguelin on the proliferation and apoptosis of ovarian cancer SKOV cells and its pos-sible mechanism.Methods Experiments were conducted from March 2019 to March 2020. After deguelin acted on SKOV cells, Cell counting kit-8 (CCK-8) was used to detect cell proliferation, flow cytometry was used to detect cell apoptosis, and Western blotting was used to detect cyclin-dependent kinase inhibitor 1 (P21), Cyclin D1, B-lymphoma-2 (Bcl-2), Bcl-2-related X protein (Bax), Kruppel-like factor 15 (KLF15) protein expression levels were detected, real-time quantitative PCR (RT-qPCR) ) to detect the expression level of KLF15 mRNA. After transfecting KLF15 overexpression vector to up-regulate KLF15 expression in SKOV cells, the same methods as above were used to observe the effect of up-regulating KLF15 expression on SKOV cell proliferation, apoptosis and the expression of Cy-clinD1, P21, Bcl-2 and Bax protein.Results After deguelin acted on SKOV3 cells, cell activity (0.49±0.04, 0.39±0.03, 0.30±0.02), pro-tein expression levels of CyclinD1 (0.53±0.05, 0.45±0.05, 0.21±0.03) and Bcl-2 (0.40±0.04, 0.24±0.02) , 0.16±0.01) were decreased (P< 0.05), apoptosis rate [(6.63±0.69)%, (12.71±1.06)%, (20.27±2.25)%], protein expression levels of P21 (0.23±0.02, 0.46±0.04, 0.74±0.03), Bax (0.20±0.02, 0.36±0.02, 0.55±0.05) protein expression level increased, KLF15 mRNA and protein expression levels increased(P<0.05). After up-regulating KLF15 expression, SKOV3 cell activity, protein expression levels of CyclinD1 and Bcl-2 were decreased (P <0.05), apoptosis rate, protein expression levels of P21 and Bax were increased (P<0.05). After down-regulating the expression of KLF15, the SKOV3 cell activity, CyclinD1 and bcl-2 protein expression levels of cells increased by treating deguelin (P<0.05), the apoptosis rate, P21 and Bax protein expression levels were decreased (P<0.05).Conclusions Deguelin can inhibit the proliferation of ovarian cancerSKOV3 cells and induce the apoptosis of SKOV3 cells, and its mechanism is related to the up-regulation of KLF15 expression in the cells.
