      目的分析食管癌组织中微 RNA-634(miR-634)和锌指蛋白 ZBTB20的表达水平,并分析二者是否与食管癌发生发展及预后相关。方法选取 2018年 1月至 2019年 4月湖北医药学院附属国药东风总医院接受手术的食管癌病人 102例,从病人根治切除术手术标本切取癌组织及癌旁正常组织。实时荧光定量 PCR(qRT-PCR)检测 miR-634、ZBTB20 mRNA相对表达水平。 Pearson相关性分析食管癌组织中 miR-634与 ZBTB20 mRNA表达水平的相关性。 Kaplan-Meier生存曲线分析 miR-634、 ZBTB20与预后的关系。 Cox回归分析食管癌病人预后危险因素。结果与癌旁正常组织( 1.02±0.06、1.01±0.05)相比,食管癌组织中 miR-634低表达( 0.69±0.15)(P<0.05)ZBTB20 mRNA高表达( 1.64±0.26)(P<0.05)。 Pearson相关性分析显示, miR-634、 ZBTB20 mRNA在食管癌病人的组织样本中表达,水平负相关( r=.0.65,P<0.05);两者均与 TNM分期、淋巴结转移、组织分化程度相关( P<0.05)。 Kaplan-Meier法显示, miR-634高表达组( 64.71%)无进展生存率较低表达( 29.41%)高( χ2=14.41,P<0.001), ZBTB20低表达组( 62.75%)无进展生存率较高表达组( 31.37%)高( χ2=9.48,P=0.002)。多因素 Cox回归分析表明, TNM分期(Ⅲ)、淋巴结转移、 miR-634低表达、 ZBTB20高表达是影响食管癌病人不良预后的危险因素( P<0.05)。结论食管癌病人癌组织中 miR-634表达水平降低, ZBTB20表达水平升高,两者与 TNM分期、淋巴结转移、组织分化程度及预后密切相关。

      Objective To analyze the expression levels of micrornA-634 (miR-634) and zinc finger protein ZBTB20 in esophagealcancer tissues,and to analyze their correlation with the occurrence,development and prognosis of esophageal cancer.Methods A basic study was conducted on 102 patients with esophageal cancer who underwent surgery in Dongfeng General Hospital of Sinopharm,HubeiMedical College from January 2018 to April 2019, and the cancer tissues and adjacent normal tissues were cut from the surgical speci.mens of patients undergoing radical resection. The relative expression levels of miR-634 and ZBTB20 mRNA in esophageal cancer tis. sues and adjacent normal tissues were detected by real-time fluorescentquantitative PCR (qRT-PCR). Pearson correlation was applied to analyze the correlation between miR-634 and ZBTB20 mRNA expression levels in esophageal cancer tissues.Kaplan-Meier survival curve was applied to analyze the relationship between miR-634, ZBTB20 and prognosis. Cox regression analysis was applied to analyzethe risk factors affecting the prognosis of patients with esophageal cancer.Results Compared with adjacent normal tissues (1.02±0.06,1.01±0.05), the expression of miR-634 was lower (0.69±0.15) (P<0.05) and ZBTB20 mRNA was higher (1.64±0.26) (P<0.05) in esopha. geal cancer tissues. Pearson correlation analysis showed that the expression levels of miR-634 and ZBTB20 mRNA were negatively cor. related in tissue samples of patients with esophageal cancer (r=.0.65, P<0.05). Both were correlated with TNM stage, lymph node me. tastasis and tissue differentiation (P<0.05). Kaplan-Meier method showed that the high miR-634 expression group (64.71%) had higher progression-free survival than the low Mir-634 expression group (29.41%) (χ2=14.41, P<0.001), and the low ZBTB20 expression group(62.75%) had higher progression-free survival than the low ZBTB20 expression group (31.37%) (χ2=9.48, P=0.002). Multivariate Cox regression analysis showed that TNM stage (Ⅲ), lymph node metastasis, low expression of miR-634 and high expression of ZBTB20 were risk factors for poor prognosis of patients with esophageal cancer (P<0.05).Conclusion The expression level of miR-634 is de. creased, and the expression level of ZBTB20 is increased in the cancer tissues of patients with esophageal cancer, and the two are close.ly related to TNM stage, lymph node metastasis, degree of tissue differentiation and prognosis.
