目的:探讨天丹通络胶囊(Tiandan Tongluo Capsules,TD)调控LKB1/AMPK信号通路对急性脑梗死神经炎症的影响。方法:将48只Sprague-Dawley大鼠随机分为sham组、model组、天丹通络胶囊低剂量组(TD-L组)和天丹通络胶囊高剂量组(TD-H组)。采用金属丝闭塞法建立大鼠脑梗死模型。术后第7天进行动物行为学分析,观察天丹通络胶囊对细胞的病理及神经保护作用。ELISA检测炎性因子的表达。TUNEL染色分析细胞凋亡。免疫组化和Western blot分析LKB1/AMPK信号通路活性。结果:与sham组相比,model组的Zea-Longa评分显著升高,而TD-L组和TD-H组的评分则明显低于model组。ELISA分析结果表显示,model组促炎因子IL-1β和TNF-α的水平显著升高,抗炎因子IL-10的表达减少,而TD-L组和TD-H组IL-1β和TNF-α的水平显著降低,抗炎因子IL-10的表达提高。此外,model组TUNEL阳性细胞数量明显升高,而TD-L组和TD-H组的TUNEL阳性细胞数量则明显低于model组。TD-L组和TD-H组cleaved caspase-3和cleaved caspase-9表达相较于model组显著降低。与sham组相比,model组LKB1和AMPK的相对蛋白表达水平显著降低。与model组相比,TD-L组和TD-H组的LKB1和AMPK水平显著升高。结论:天丹通络胶囊可抑制脑梗死大鼠神经细胞凋亡,发挥保护作用,这可能与LKB1/AMPK信号通路有关。 |

[Objective] To explore the effect of Tiandan Tongluo Capsules(TD) regulating LKB1/AMPK signaling pathway on neuroinflammation in acute cerebral infarction. [Method] Forty-eight Sprague-Dawley rats were randomly divided into sham group, model group, Tiandan Tongluo Capsules low-dose group (TD-L group) and Tiandan Tongluo Capsules high-dose group (TD-H group). The rat model of cerebral infarction was established by wire occlusion. Animal behavior analysis was performed on the 7th day after surgery to observe the pathological and neuroprotective effects of Tiantan Tongluo tablet on cells. The expression of inflammatory factors was detected by ELISA. Apoptosis was analyzed by TUNEL staining. The activity of LKB1/AMPK signaling pathway was analyzed by immunohistochemistry and Western blot. [Result] Compared with sham group, Zea-Longa scores of model group were significantly increased, while scores of TD-L and TD-H groups were significantly lower than those of model group. The results of ELISA analysis showed that the levels of pro-inflammatory factor IL-1β and TNF-α were significantly increased in model group, and the expression of anti-inflammatory factor IL-10 was decreased, while the levels of IL-1β and TNF-α were significantly decreased in TD-L and TD-H groups, and the expression of anti-inflammatory factor IL-10 was increased. In addition,. The number of TUNEL positive cells in model group was significantly increased, while the number of TUNEL positive cells in TD-L and TD-H groups was significantly lower than that in model group. The expression of cleaved caspase-3 and cleaved caspase-9 in the TD-L and TD-H groups was significantly lower than that in the model group. Compared with sham group, the relative protein expression levels of LKB1 and AMPK in model group were significantly decreased. Compared with model group, LKB1 and AMPK levels in TD-L and TD-H groups were significantly increased. [Conclusion] Tiandan Tongluo Capsules can inhibit nerve cell apoptosis in rats with cerebral infarction and play a protective role, which may be related to LKB1/AMPK signaling pathway. |
