      目的:探讨CircAPLP2通过靶向miR-455-3p对子宫内膜癌(endometrial carcinoma,EC)恶性生物学行为的作用。方法:从TCGA-UCEC中下载miRNA表达数据,筛选目标miRNA。Starbase数据库预测miRNA上调调控分析。利用qRT-PCR检测miR-455-3p和CircAPLP2 mRNA的表达。蛋白质免疫印迹法分析细胞周期相关蛋白和Notch信号通路相关蛋白的表达。利用生信预测以及双荧光素酶报告基因分析以验证miR-455-3p与CircAPLP2的靶向关系。克隆形成实验检测EC细胞的增殖,流式细胞术检测EC细胞周期进程,划痕愈合实验测定EC细胞的迁移,Transwell分析法评估EC细胞的侵袭能力。结果:miR-455-3p在EC细胞中低表达。过表达miR-455-3p抑制EC细胞的增殖、迁移和侵袭,敲低miR-455-3p的表达则增强EC细胞的增殖、迁移和侵袭。此外,CircAPLP2作为miR-455-3p的分子海绵。敲低CircAPLP2和PBK则能逆转过表达miR-101-5p对EC细胞增殖和转移的抑制作用。结论:CircAPLP2通过靶向miR-455-3p激活Notch信号通路促进EC的增殖和转移,这可能为认识EC恶性进展提供了一个新的途径。
英文摘要:
      [Objective] To investigate the effect of CircAPLP2 on the malignant biological behavior of endometrial carcinoma ( EC ) by targeting miR-455-3p. [Method] The miRNA expression data were downloaded from TCGA-UCEC to screen the target miRNA. Starbase database predicted miRNA up-regulation regulation analysis. The expression of miR-455-3p and CircAPLP2 mRNA was detected by qRT-PCR. Western blotting was used to analyze the expression of cell cycle-related proteins and Notch signaling pathway-related proteins. Bioinformatics prediction and dual luciferase reporter gene analysis were used to verify the targeting relationship between miR-455-3p and CircAPLP2. Clone formation assay was used to detect the proliferation of EC cells. Flow cytometry was used to detect the cell cycle progression of EC cells. Scratch healing assay was used to detect the migration of EC cells. Transwell assay was used to evaluate the invasion ability of EC cells. [Result] MiR-455-3p was lowly expressed in EC cells. Overexpression of miR-455-3p inhibited the proliferation, migration and invasion of EC cells, while knockdown of miR-455-3p enhanced the proliferation, migration and invasion of EC cells. In addition, CircAPLP2 acts as a molecular sponge for miR-455-3p. Knockdown of CircAPLP2 and PBK reversed the inhibitory effect of overexpression of miR-101-5p on the proliferation and metastasis of EC cells. [Conclusion] CircAPLP2 promotes the proliferation and metastasis of EC by targeting miR-455-3p to activate Notch signaling pathway, which may provide a new way to understand the malignant progression of EC.
