      摘要:目的 建立土家族药物烂泥巴树HPLC指纹图谱,并进行化学模式识别分析评价烂泥巴树药材质量。方法 采用Kromasil 100-5 C18色谱柱(250mm*4.6m,5μm),以乙腈(A)-0.2% 磷酸水溶液(B)为流动相,梯度洗脱,流速1.0 mL/min,检测波长220 nm,柱温30 ℃,进样量10 μL。通过《中药色谱指纹图谱相似度评价系统》进行相似度评价,并结合聚类分析(CA)、主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)对不同产地烂泥巴树进行综合分析。结果 建立了土家族药物烂泥巴树HPLC指纹图谱,共提取17个共有峰,并指认出3号峰为紫丁香苷,4号峰为绿原酸,9批烂泥巴树指纹图谱与对照指纹图谱的相似度均大于0.9,CA将药材分为2类,PCA、OPLS-DA结果与CA一致,并筛选出11个差异标志物。结论 建立的土家族药物烂泥巴树HPLC指纹图谱相似度好,结合化学模式识别,可以为不同产地烂泥巴树质量控制提供参考。

      Abstract:Objective Establish an HPLC fingerprint of the Tujia ethnic medicine,Torricellia Angulata,and conduct chemical pattern recognition analysis to evaluate the quality of Torricellia Angulata medicinal material. Methods Using Kromasil 100-5 C18 chromatographic column (250mm * 4.6m, 5μm),using acetonitrile (A) -0.2% phosphoric acid aqueous solution (B) as the mobile phase,gradient elution,flow rate of 1.0 mL/min,detection wavelength of 220 nm,column temperature of 30 ℃,and injection volume of 10 μL.Perform similarity evaluation using the Traditional Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System,and combine cluster analysis (CA),principal component analysis (PCA),and orthogonal partial least squares discriminant analysis (OPLS-DA) to conduct comprehensive analysis on different production areas of Torricellia Angulata. Results Establish an HPLC fingerprint of the Tujia ethnic medicine,Torricellia Angulata.A total of 17 common peaks were extracted,and identify the third peak as syringin and the fourth peak as chlorogenic acid.The similarity between the fingerprint of 9 batches of Torricellia Angulata and the control fingerprint was greater than 0.9.CA classified the medicinal materials into 2 categories, and the PCA and OPLS-DA results were consistent with CA.Eleven differential biomarkers were screened.Conclusion The established HPLC fingerprint of the Tujia ethnic medicine Torricellia Angulata has good similarity,and combined with chemical pattern recognition,it can provide reference for quality control of Torricellia Angulata in different production areas.
