      目的分析长链非编码 RNA(lncRNA)TPT1-AS1在非小细胞肺癌中的表达及其与临床病理特征的关系。方法收集 2014年 7月至 2016年 9月陕西省核工业二一五医院非小细胞肺癌标本及癌旁组织各 100例,采用实时荧光定量 PCR(qRT-PCR)检测非小细胞肺癌标本、癌旁组织中 lncRNA TPT1-AS1表达情况,分析 lncRNA TPT1-AS1表达水平与非小细胞肺癌病人临床病理特征及预后的关系。结果与癌旁肺组织相比, lncRNA TPT1-AS1在肺癌组织的表达显著升高( P<0.05)。肺癌组织中 lncRNA TPT1-AS1表达与 TNM分期、淋巴转移有相关性(P<0.05)。与 lncRNA TPT1-AS1低表达组生存率( 41.46%)相比, lncRNA TPT1-AS1高表达组非小细胞肺癌病人生存率(10.17%)显著降低(χ2=13.376,P<0.05)。多因素分析显示, lncRNA TPT1-AS1表达和 TNM分期是影响非小细胞肺癌病人预后的独立危险因素( HR=1.954,95% CI:1.545-2.473,P<0.05;HR=2.013,95% CI:1.5912.547,P<0.05)。结论 lncRNA TPT1-AS1与非小细胞肺癌的发生发展及预后密切相关,可能作为临床预后判断的参考指标。
英文摘要:
      Objective To analyze the expression of long non-coding RNA (lncRNA) TPT1-AS1 in non-small cell lung cancer (NSCLC) and its relationship with clinicopathological features.Methods One hundred samples of NSCLC and 100 adjacent tissueswere collected in No.215 Hospital of Shanxi Nuclear Industry from July 2014 to September 2016, the expression of lncRNA TPT1-AS1 in NSCLC and adjacent tissues was detected by real-time fluorescent quantitative PCR (qRT-PCR), and the relationships between the expression of lncRNA TPT1-AS1 and the clinicopathological features, prognosis of NSCLC were analyzed.Results The expression of lncRNA TPT1-AS1 in lung cancer tissue was significantly higher than that in paracancerous lung tissue (P<0.05). The expression of lncRNA TPT1-AS1 was correlated with TNM stage and lymph metastasis (P<0.05). Compared with the survival rate (41.46%) in the low expression group of lncRNA TPT1-AS1, the survival rate (10.17%) in the high expression group of lncRNA TPT1-AS1 was significantly lower (χ2=13.376, P<0.05). Multivariate analysis showed that the expression of lncRNA TPT1-AS1 and TNM stage were independent risk factors for the prognosis of NSCLC (HR = 1.954, 95% CI:1.545-2.473, P<0.05; HR = 2.013, 95%CI:1.591-2.547, P<0.05).Conclu? sion lncRNA TPT1-AS1 is closely related to the occurrence, development and prognosis of NSCLC, which may be a reference index for clinical prognosis.
