      目的探讨迷迭香酸联合槲皮素对人肝癌 HepG2细胞增殖、侵袭力的影响,并探讨其可能的机制。方法以不同浓度迷迭香酸(12.5、25.0、50.0和100.0 μmol/L)和槲皮素(12.5、25.0、50.0和100.0 μmol/L)单药及二者联合用药处理 HepG2细胞 36 h,用四甲基偶氮唑盐微量酶反应比色法(MTT法)检测细胞增殖,划痕实验检测细胞迁移能力,蛋白质印迹法检测凋亡相关蛋白 E-钙黏蛋白(E- cad)及N-钙黏蛋白(N-cad)的表达情况。结果槲皮素和迷迭香酸单药均能使细胞活力降低,二者联合用药对细胞活力抑制率更加显著,且呈剂量依赖性( P<0.05)。 25 μmol/L迷迭香酸组及25 μmol/L槲皮素组划痕愈合距离分别为( 11.35±2.37)mm、(11.46±

      Objective To investigate the effect of combined use of quercetin and rosmarinic acid on proliferation and apoptosis of HepG2 cells,and to explore the possible mechanism.Methods HepG2 cells were treated with different concentrations of quercetin(12.5,25.0,50.0,100.0 μmol/L)and rosmarinic acid(12.5,25.0,50.0,100.0 μmol/L)alone or in combination for 36 hours.Cell proliferation was assayed by MTT method.The ability of cell migration was studied by wound healing test and expressions of E-cadprotein and N-cad were measured by Western blot.Results Both quercetin and rosmarinic acid reduced cell viability,and the com- bination of the two drugs had a more significant inhibition on cell viability in a dose-dependent manner(P<0.05).The wound heal- ing distances of the 25 μmol/L rosmarinic acid group and 25 μmol/L quercetin group were(11.35±2.37)mm and(11.46±3.86) mm,respectively,both of which inhibited cell migration;the wound healing distance in the combined medication group was(4.36±0.56)mm,with more significant inhibition on cell migration(F=6.73,P=0.003).The relative expression of E-cad protein in the quercetin group and rosmarinic acid group were(1.02±0.10)and(1.25±0.14),and the relative expression of N-cad protein was(0.98±0.05)and(0.73±0.02).In contrast,the combination of the two drugs significantly increased E-cad[(1.42±0.06),F=5.28, P=0.02]and downregulated N-cad[(0.61±0.07),F=3.28,P=0.03]expression.Conclusion Quercetin combined with rosmarin-ic acid can synergistically inhibit cell proliferation and migration probably by up-regulating the expression of E-cad and down-regu-lating N-cad gene expression.
