      目的探讨富含亮氨酸重复序列的 G蛋白偶联受体 6(LGR6)对人结肠癌 SW480细胞增殖和侵袭的作用及机制。方法在结肠癌细胞株 SW480细胞中构建低表达 LGR6的实验组、阴性对照组和空白对照组,用实时荧光定量核酸扩增检测(qPCR)和蛋白质印迹法( Western Blot)验证 LGR6 mRNA和蛋白的表达水平。用 MTT法检测细胞增殖活性,用 Transwell小室法检测细胞的侵袭能力,用 Western Blot检测 Wnt/β-连环蛋白( β catenin)信号通路相关蛋白表达水平。结果实验组、阴性对照组和空白对照组 LGR6 mRNA表达水平分别为( 0.42±0.08)、(1.12±0.08)、(1.13±0.31),转染 LGR6 siRNA的 SW480细胞 LGR6 mRNA和蛋白表达水平显著降低(均 P < 0.05);转染后 24、48、72 h时实验组细胞增殖活性[( 0.24±0.04)、(0.27±0.06)、(0.45±0.08)、(0.63±0.09)]显著低于阴性对照组[( 0.22±0.03)、(0.40±0.05)、(0.92±0.07)、(1.32±0.11)]和空白对照组[( 0.26±0.04)、(0.42±0.07)、(0.94±0.09)、(1.21±0.12)均 P < 0.05]。下调 LGR6表达可促进结肠癌细胞 Bax和 Cleaved caspase 3蛋白的表达,并抑制结肠癌细胞 β catenin和 c-Myc蛋白,的表达(均 P < 0.05)。结论下调 LGR6可能通过调控 Wnt/β-catenin信号通路抑制结肠癌 SW480细胞的增殖和侵袭,并促进结肠癌 SW480细胞的凋亡。

      Objective To investigate the effect of G protein-coupled receptor 6 rich in leucine repeat sequence (LGR6) on proliferation and invasion of human colon cancer SW480 cells and its mechanism.Methods The low expression of LGR6 experimental group,negative control group and blank control group were constructed in colon cancer cell line SW480 cells. The expression levels of LGR6mRNA and protein were measured by qPCR and Western Blot, respectively. The proliferation activity of the cells was detected by MTTassay, the invasion ability of the cells was detected by Transwell chamber method, and the expression level of Wnt/β catenin signalingpathway-related proteins was detected by Western Blot.Results The expression levels of LGR6 mRNA in the experimental group, negative control group and blank control group were (0.42±0.08),(1.12±0.08),(1.13±0.31), respectively. The LGR6 mRNA and protein expression levels of SW480 cells transfected with LGR6 siRNA were significantly reduced (all P < 0.05); The cell proliferation activity ofthe experimental group (0.24±0.04), (0.27±0.06), (0.45±0.08), (0.63±0.09) at 24 h, 48 h, and 72 h after transfection was significantlylower than that of the negative control group (0.22±0.03),(0.40±0.05), (0.92 ±0.07), (1.32 ± 0.11) and the blank control group (0.26 ±0.04),(0.42 ± 0.07),(0.94 ± 0.09),(1.21 ± 0.12), all P < 0.05). down-regulation of LGR6 expression promoted the expression of Bax andCleaved caspase 3 proteins in colon cancer cells, and inhibited the expression ofβ catenin and c-Myc proteins in colon cancer cells (all P < 0.05).Conclusion Downregulation of LGR6 may inhibit the proliferation and invasion of colon cancer SW480 cells and promoteapoptosis by regulating Wnt/β-catenin signaling pathway.
